Purpose: Filtrating 2 components of 5 effective components which are total saponins, polysaccharide, total flavonoids of Astragalus, polysaccharide of Angelica and ferulic acid in Angelica sinensis decoction of supplenmenting blood by the methods of combinations of bone marrow cell culture and animal experiments. Then decide the best ratio of the 2 selected components and lay the foundation of the components proportion of Angelica sinensis decoction of supplenmenting blood.Methods: Firstly, we culture bone marrow of SD rats and detect cell proliferation activity of the cultured bone marrow with MTT methods in order to select the best 2 components in irritating bone marrow’s proliferation function among Angelica sinensis decoction of supplementing blood’s 5 effective components. Then we make up 6 different ratios’ fluids with the 2 selected components which are 10:0, 8:2, 6:4, 4:6, 2:8, and 0:10, and gastric the rats model which is deficient in Qi and Blood. After gastric the model rats, calculate the number of hemocyte and test the content of EPO,SCF,IL-3,IL-2,IL-6,G-CSF,GM-CSF in blood, and decide the best proportion of the 2 selected components.Results: According to the bone marrow results, we summarize that the middle and high dosage groups of Angelica polysaccharide and high dosage group of Astragalus total saponins have significantly accelerated the proliferation of bone marrow and the high dosage group of Astragalus polysaccharide and ferulic acid have the tendency of increase proliferation to the bone marrow, but no statistical significance.According to the rats experiment, thymus index of Angelica sinensis decoction of supplementing blood increase significantly, while the other groups have no statistical significance, even though there are little damage seen from microscope observation. All gastric groups have no significant difference to the spleen index and kidney index. While from the pathogenesis results, the gastric groups can irritate spleen to generate blood, especially in the group of 6:4 in Astragalus total saponins and Angelica polysaccharide. And there is no difference in spleen’s pathogenesis’ slice. And this group can significantly increase the number of hemocyte and hemoglobin. So as to EPO. It has no function in increase the number of leukocyte, but has the tendency of decreasing its number. The 6:4 group can also decrease the content of IL-2 and IL-6. The group 2:8 and 0:10 in Astragalus total saponins and Angelica polysaccharide both can decrease the content of IL-3 and SCF. All gastric groups have the tendency in decreasing GM-CSF and G-CSF, but no statistical significance.Conclusion: when the proportion of Astragalus total saponins and Angelica polysaccharide is 6:4, it have best function in increasing the number of hemocyte.
