Reversal Effect And Its Mechanism Of ShenMai Injection On Adriamycin Resistance In Breast Cancer

Shenmai injection is a traditional Chinese medicine compound injection refined from red ginseng and Ophiopogon japonicus.It has a wide range of pharmacological effects.Ginsenoside and Ophiopogon saponin are the main active ingredients.To investigate the reversal effect of Shenmai Injection in Adriamycin resistant breast cancer cell line,named MCF-7/ADR,which was used in in vivo and in vitro experiments.An UHPLC method was developed to achieve simultaneous quantitation of 11 main ginsenosides(Rg1,Re,Rf,20(S)-Rh1 20(S)-Rg2,Rb1,Rc,Rb2,Rb3,Rd,20(S)-Rg3)in Shenmai Injection.Meanwhile,multiple batches of Shenmai Injection were examined.Used MTT assy and Annexin V/PI staining,the results showed that Shenmai Injection was nontoxic to breast cancer cells at 1 mg/mL;The results suggested that MCF-7/ADR cells were approximately 14-fold more resistant to Adriamycin than the MCF-7 cells.;After MCF-7/ADR cells treated with Shenmai injection,the toxicity of Adriamycin to MCF-7/ADR cells was strengthened significantly;Annexin V/PI staining to detect the effect of Shenmai injection combined with Adriamycin on apoptosis.The mechanism was explored using rhodamine-123 retention,HPLC-FLD,Fluorescence microscopy,Western-blotting.The results showed that SMI could inhibit the efflux function of P-gp and its expression.The MCF-7/ADR inoculated mice xenograft model was used in vivo experiment.The results indicated that Shenmai injection combined with Adriamycin could effectively inhibit the growth of tumor in xenografts mice,and the expression of P-gp in the tumor tissue decreased significantly in combined treatment group.
