Objective: Serine/threonine protein kinase is an important member of the protein kinase family.Its function is to phosphorylate serine and threonine residues in proteins.It plays a key role in regulating important cellular processes such as DNA replication,signal transduction pathways,cell proliferation,cell differentiation,cell death and tumor genesis and development.Recently,many members of the STK family have been reported to be associated with tumors.Among them,STK32 B has been found to play a key role in the metastasis and angiogenesis of oral squamous cell carcinoma,but there is no related research on expression of STK32 B in glioma.In this study,we investigated the expression of STK32 B in glioma of different grades and normal brain tissues,and further explored the correlation between the expression of STK32 B and the prognosis of glioma patients.Methods: Collecting 37 cases of glioma patients' specimens confirmed by pathologists after operation in the Department of Neurosurgery in The First Hospital of Chinese Medical University and the recording corresponding clinical and prognostic data.Taking normal brain tissue as control group,observe the expression of STK32 B in glioma samples by Western blot and immunohistochemistry.Then,analyze the correlation between the expression level of STK32 and the grade of glioma.Using immunohistochemistry to detect the protein level of STK32 B and then analyze the expression of STK32 B protein in relation to clinical pathological factors such as sex,age,tumor grade,KPS score and prognosis.Kaplan-Meier survival curve was made and COX proportional hazard model was used for multivariate analysis.Results:(1)Immunohistochemical staining showed that STK32 B expression was detected in 37 glioma specimens and 9 normal brain tissue specimens,including 9 cases of grade IV glioma,9 cases of grade III glioma,6 cases of WHO grade II glioma,6 cases of normal brain tissue,and 9 cases of grade III and IV glioma.The expression of STK32 B in tumor specimens was significantly different from that in normal brain tissues.(2)Western blot showed that STK32 B was expressed in both glioma and normal brain tissues.The expression of STK32 B in WHO grade III and IV gliomas was higher than that in normal brain tissues,and the difference was statistically significant.(3)The correlation analysis between STK32 B expression in glioma and clinicopathological characteristics showed that STK32 B expression in glioma tissue was not related to WHO grade,age,sex and tumor size.(4)The survival rate of STK32 B high expression group was significantly different from that of low expression group(P < 0.05).The validation results of database retrieval are basically consistent with our research results.Conclusions: The expression of STK32 B in glioma tissue was higher than that in normal brain tissue.The expression of STK32 B was related to the prognosis of glioma patients.
