?Objectives?To explore the proliferation and mechanism of HBD1 on neonatal umbilical cord blood CD4~+T cells.?Methods?Cord blood mononuclear cells or purified CD4~+T lymphocytes were divided into four groups:(1)negative control group:untreated;(2)HBD1 treatment group:treated with HBD1;(3)positive control group:treated with PHA(PMA/Ion);(4)co-treatment group:co-treated with HBD1 and PHA(PMA/Ion).The proliferation of CD4~+T lymphocytes was detected by CCK-8.The expression of CD80?CD86 on B cells and CD69?CD25?CD40L?CD45RO?IL-2on CD4~+T cells was detected by FACS.At the same time,the adult peripheral blood as control.?Results?For cord blood,comparing with group(1)and group(3),group(2)or group(4)promotes The proliferation of CD4~+T cells,the expression of CD69?CD25?CD40L?IL-2 and the expression of CD80?CD86,all they showed statistical differences;but There was no significant difference between the two groups in the expression of CD45RO.And There was no significant difference between the two groups in adult peripheral blood.?Conclusion?HBD1 promotes the proliferation and activation of CD4~+T cells in neonatal umbilical cord blood and promotes the secretion of CD4~+T cytokine IL-2.HBD1 may improve the immunity of newborns.The same treatment conditions,HBD1on adult peripheral blood-derived CD4~+T cells immunoregulatory.
