Interstitial cystitis(IC),also known as bladder pain syndrome(BPS),is a common disease with unclear pathogenesis and difficult to treat. It is of great importance to establish an ideal animal model of IC to investigate the pathogenesis and therapeutic strategies of IC.C57BL/6 strain mice are widely used in scientific research with many advantages including low-cost,multiple knockout strains, and well-characterized genetics. However,up to now,no effective model of IC in C57BL/6 strain mice has been made.We found that duplicate immunization with bladder homogenate can establish an experimental autoimmune cystitis(EAC)model that can simulate the pathophysiology and clinical symptoms of IC,which can be used to research the pathogenesis and therapeutic strategies of IC.The bladder pain of IC with urine frequency and urgency disturbed patients seriously while existing treatments of IC are not satisfactory. In our previous study, neurokinin receptor-1(NK1R)inhibitor(aprepitant)could relieve the inflammation of severe non-bacterial cystitis. We felt obliged to investigate whether blocking NK1R could treat IC like lesion or not.Our results suggested that NK1R inhibitor aprepitant could relieve the pain in the bladder area and reduce the inflammation of IC bladder. Our research may provide a new idea and method in the treatment of IC.
