Cancer has become the world's second leading cause of death,death caused by cancer is second only to cardiovascular and cerebrovascular diseases.And among all kind of cancer types,Lung cancer is the most prevalent cancer types.It is of great significance for the diagnosis and treatment of lung cancer in the background of severe haze in China today.Early diagnosis and treatment of lung cancer can greatly improve the survival rate of patients,but how to achieve early diagnosis of lung cancer is always a difficult medical profession.In this study,dual-modal and dual-targeting nano-probe preparation,combined with SPECT/MRI imaging technology for early diagnosis of lung cancer were involved.The RGD peptide with?v?3 targeting property and Ge11 peptide with EGFR targeting property were conjugated to the surface of ferrous tetroxide nanoparticles by modifying the nanoparticles.And its targeting properties,cytotoxicity and stability were verified.Then 99m Tc nuclide was labeled on the surface of the probe.The imaging and early diagnosis of the material were evaluated by SPECT and MRI imaging.Ability as T1 and T2contrast agent of iron oxide nanoparticles were then evaluated by T1 and T2imaging of MRI,respectively.There are four main parts:?v?3 intergrin and EGFR dual targeting,?v?3 single-targeting,EGF receptor single-targeting nanoprobe construction;three different probes in vitro performance evaluation;probe in vivo performance evaluation and Evaluation of T1/T2 magnetic resonance imaging of iron nanoparticles.The main results and conclusions are as follows:?1?The superparamagnetic Fe3O4nanoparticle probe with good stability,low toxicity and targeting receptor of?v?3 and EGF receptor was successfully prepared.?2?SPECT and MRI imaging of?v?3 single target,EGF receptor single target,and?v?3 receptor and EGF receptor dual targeting targeting probe were performed respectively,which proved the superiority of dual target targeting Diagnostic performance.?3?The magnetic resonance imaging properties of ultrafine-sized Fe3O4 nanoparticles were studied.It was proved that the iron nanoparticles had better T2 contrast effect.
