Objective To observe the effect of 18?-sodium glycyrrhetinic acid on the expression of TNF-? and TGF-?1 in nasal mucosa of allergic rhinitis rats,and to explore the intervention mechanism of 18?-sodium glycyrrhetinic acid on allergic rhinitis.Methods 100 SPF-level wistar rats were randomly divided into control group,allergic rhinitis group,budesonide group,18?-sodium glycyrrhetinic acid low-dose group(20 mg/kg)and18?-sodium glycyrrhetinic acid high-dose group(40 mg/kg).The AR rat model was constructed by OVA sensitization and then given the drug intervention.The rats in each group were sacrificed after 2 and 4 weeks of intervention and animal behavioral score.Serum was taken for ELISA to detect the expression of OVA-sIgE.HE staining and immunohistochemical staining were performed on the nasal mucosa of rats.Immunofluorescence staining,RT-PCR and western-blot were used to quantify the expression of TNF-? and TGF-?1 in rat nasal mucosa.Results By the animal behavioral scores,ELISA of serum OVA-sIgE and HE staining,we found that the animal model of allergic rhinitis was successfully constructed.By immunofluorescence and immunohistochemistry,we found that TNF-? and TGF-?1 mainly existed in the columnar epithelium,vascular endothelium,gland and part of inflammatory cytoplasm of nasal mucosa.The results of western-blotting and RT-PCR showed that the expression of TNF-? and TGF-?1 in the nasal mucosa of allergic rhinitis rats were significantly higher than that in normal rats(P < 0.01).After different doses of 18?-sodium glycyrrhetinic acid or budesonide,the expression of TNF-? and TGF-?1 was significantly decreased(P < 0.01),especially in the low-dose 18?-glycyrrhetinic acid group at 4 weeks(P < 0.01),and the change is positively correlated.Conclusion Different doses of 18?-sodium glycyrrhetinic acid have an effect on allergic rhinitis,especially in the low-dose group at 4 weeks,and its mechanism may be related to the inhibition of high expression of TNF-? and TGF-?1.
