Objective: To analyze the expression of EDIL3 in bladder cancer and adjacent tissues,the expression of EDIL3 gene in different grades of bladder cancer and its correlation with vascular endothelial growth factor(VEGF),so as to provide theoretical support for clinical diagnosis and treatment of bladder cancer.Methods: 56 paraffin specimens from 2007 to 2013 in the Second Hospital of Lanzhou University and 25 specimens from 2015 to 2016 in Xianyang first people's hospital were selected.The paraffin specimens were cut into 3um thick sections continuously without interruption by routine xylene dewaxing and gradient alcohol hydration.The specimens were all inactivated by PV9001 two-step method(abiotin);the endogenous peroxidase was blocked by 3% hydrogen peroxide deionization.Water was incubated at room temperature for 15 minutes,PBS was rinsed three times,3 minutes per time;absorbent paper was used to absorb PBS and drip anti-PBS(the best dilution ratio was pre-determined by experiment: EDIL3 1/200 dilution concentration,the concentration of VEGF 1/100 dilution concentration),incubation time was 0.5h37?.DAB solution was used for color rendering,then tap water was used to rinse,re-dye,dehydrate,transparent and seal the tablets.PBS was used instead of primary antibody in the negative control test.The results under light microscope were consistent with other operations.Results: 1.EDIL3 protein was found in cells and extracellular matrix,and brown granules were found in positive cells and extracellular matrix.The expression of EDIL3 protein in bladder cancer was significantly higher than that in adjacent tissues(P < 0.05).2.The high expression rates of EDIL3 protein in primary,secondary and tertiary bladder cancer tissues were 19.05%,60.00% and 70.59%,respectively.It can be seen that the expression level of EDIL3 protein increased with the increase of malignant grade of bladder cancer tissues.The results showed that there were significant differences between the first and the second grades(2 = 6.361,P < 0.05);significant differences between the first and the third grades(2 = 10.238,P < 0.05);and no significant differences between the second and the third grades(2 = 0.396,P > 0.05).3.There were 38 cases(67.86%)with high expression of vascular endothelial growth factor gene and 18 cases(32.14%)with low expression of vascular endothelial growth factor gene in 56 cases of bladder cancer.Conclusion: 1.The high expression of EDIL3 gene plays an important role in the occurrence and development of bladder cancer.At the same time,the invasion and metastasis of bladder cancer are closely related to EDIL3 gene.2.There is a positive correlation between the expression of EDIL3 gene and the expression of vascular endothelial growth factor.The occurrence,development and angiogenesis of bladder cancer are important.3.Detection of EDIL3 gene expression in bladder urothelial carcinoma may provide theoretical guidance for clinical diagnosis and prognosis.
