Objective: To investigate the expression and distribution and the significance ofaquaporin1(AQP1) and aquaporin3(AQP3) in the bladder urothelial carcinoma tissue,and relalationship of its and between histological grade and pathological stage.To study itscorrelation with recurrence of bladder cancer. Methods: Geting specimen from60casesof bladder urothelial carcinoma and30cases of normal bladder tissue, Geting specimenfrom bladder cancer tissue of15patients after transurethral resection of urinary bladdercancer and in recurrent tissue of15patients, To detect the expression of1,3aquaporins inbladder urothelial carcinoma and normal bladder tissue with immuno-histochemistry S-Pmethod, Western blot, and analyize its histological grades、pathological phases andrecurrence. And investigate the mechanism and significance of AQP1and AQP3inbladder cancer occurrence, development and recurrence. Results: The result ofimmunohistochemistry (IHC) and Western blot were basically the same; The AQP1of thebladder urothelial carcinoma and normal bladder tissue mainly expresses at microvascularendothelial cell membrane and cytoplasm, expression of the bladder urothelial carcinomawas significantly higher thanthe normal bladder tissue, following with the increasing ofthe histological grade and pathological stage of the bladder cancer, the expression of theAQP1increased significantly (P<0.05). AQP3AQP3expresses at the membrane andcytoplasm of the carcinoma cell of the bladder urothelial carcinoma, has small amount ofexpression in normal bladder tissue, and more widely distributed, besides the expressionof the protein increses as well, the difference was statistically significant (P<0.05). Theresults of this study show that the expression of aquaporin1and3increased in afterrelapse compared with the amount of the increase before relapse, and recurrence of highlevels of protein expression before, easy to relapse, and there is a negative correlationbetween amount of increasing protein expression and the recurrence time of following-up. Conclusion: the present study demonstrated that the expression of the AQP1and AQP3inthe bladder urothelial carcinoma tissue was significantly increased. The expression of theAQP1and AQP3is connected with the histological grade and pathological staging ofbladder cancer, and it plays an important role in the formatting of bladder cancer. It haspotential guide significance in the prediction of the invasive, staging and clinical guide ofbladder cancer. The the amount of the expression of aquaporin1and3apparentlyincreased in recurrence, the increasing amount and primary protein expression havenegative correlation with the recurrence time of following-up, AQP1, AQP3can be one ofthe effective indicators which is used to detect the early recurrence of bladdercancer.However, the specificity and sensitivity needs further research.
