Purpose To investigate the significance of LPA(lysophosphatidic acid)in serum in early diagnosis of epithelial ovarian cancer by detecting the plasma level of 80 patients with epithelial ovarian tumor.To detect the expression of EDG4(endothelial differentiation gene4)and PDGF-BB(Platelet-derived growth factor-BB)in 52 ovarian cancer patients and 13 normal healthy women,and to investigate the relationship between them as well as the clinicopathological features of epithelial ovarian cancer,so that we can understand the pathogenesis of epithelial ovarian cancer much better.Methods : In this paper,the level of serum expression of hemolysis phosphatidate(LPA)in patients with epithelial ovarian tumor was determined by ELISA method.The significance of early diagnosis of epithelial ovarian cancer by LPA was discussed compared with that of CA125 in the same patient.Immunohistochemistry was used to detect the expression of EDG4 and PDGF-BB in normal ovarian tissues and epithelial ovarian cancer tissues.Results:(1)LPA levels were significantly increased in serum in patients with ovarian epithelial carcinoma at various stages,and the differences were statistically significant compared with healthy women and benign epithelial ovarian tumors(P<0.001).And LPA levels were also significantly increased in the serum of stage I ovarian cancer patients,and the LPA levels in the serum of stage II-IV ovarian cancer patients were comparable,and the difference was not statistically significant(P >0.05).The level of CA125 was only slightly elevated in the serum of stage I ovarian cancer patients,and there was no statistical difference between healthy normal women and benign epithelial tumors of ovaries(P>0.05).Therefore,LPA may be used as a serological indicator for early diagnosis of ovarian cancer.(2)The expression of EDG4 and PDGF-BB increased with the changes of normal ovarian tissues and malignant epithelial ovarian cancer tissues,and the differences were statistically significant(P<0.05).(3)The positive expression of EDG4 and PDGF-BB increased with the International Federation of Gynecology of Epithelial Ovarian cancer(FIGO)staging.(4)The positive expression of EDG4 and PDGF-BB had nothing to do with the age of the patient;It was also independent of the histopathological type of the patient(P > 0.05).(5)And there was a positive correlation between EDG4 and PDGF-BB in epithelial ovarian cancer(P<0.05,r=0.075).Conclusion 1.LPA levels increased significantly in the serum of patients with epithelial ovarian cancer,LPA levels increased significantly in the serum of stage I ovarian cancer compared to CA125,It suggests that LPA is more valuable in early ovarian cancer diagnosis.2.The high expression of EDG4 is related to the occurrence,development and metastasis of epithelial ovarian cancer.3.The high expression of PDGF-BB is related to the occurrence,development and metastasis of epithelial ovarian cancer.4.The combined detection of EDG4 and PDGF-BB can serve as a reference index for the diagnosis and prognosis of ovarian cancer,and provide a theoretical basis for the molecular targeted therapy of individualized epithelial ovarian cancer patients in the future.
