Background:Antisocial Personality Disorder(ASPD)is a universal psychological behavior model that ignores or infringes the rights of others.It has the characteristics of high crime rate,high aggression,and often combined with other mental disorders,which brings great harm to social economy and public security.The cause of ASPD is still unclear.Previous studies on ASPD have shown that its pathogenesis is affected by both environmental and genetic factors.Objective:To understand the characteristics of ASPD in male violent criminals,to explore the relationship between NOS1AP gene polymorphism and ASPD,and the interaction between genes and environment,in order to provide some evidence for the pathogenesis of ASPD.Methods:Taking male prisoners of violent crimes in a prison in Nanjing as the research object,the anti-social subscale?3 in PDQ-4 was used as a positive screening criterion,and it was divided into ASPD group(166 subjects)and non-ASPD group(257 subjects).Questionnaire survey was used to collect two groups of general demographic data,family status data,material use data and psychological related data.5 ml of peripheral venous blood was taken and DNA was extracted.4 SNP(rs4145621,rs348624,rs945713 and rs1415263)of NOS1AP was genotyped by MassARRAY nucleic acid mass spectrometry.Compare the genotype frequency and allele frequency of the two groups,assess the impact of the interaction between NOS1AP gene and childhood trauma on ASPD.Result:1.There was significant difference in history of suicide(?~2=5.075,P=0.024),history of violations(?~2=11.325,P=0.001),history of prison sentences(?~2=9.049,P=0.003),history of smoking(?~2=8.305,P=0.004)and history of drug abuse(?~2=30.029,P<0.001)in ASPD group and non-ASPD group.2.The difference between the two groups was significant in the father's parenting style(?~2=17.624,P=0.001),maternal parenting style(?~2=23.253,P<0.001)and parental relationship(?~2=12.344,P=0.006).3.The differences between the two groups in general mental health(t=3.076,P=0.002),anxiety(t=6.862,P<0.001),depression(t=7.203,P<0.001)and idea of suicide were significant;total score of impulsive(t=9.114,P<0.001)and the three dimensions scores of attention impulsivity(t=7.066,P<0.001),motor impulsivity(t=8.235,P<0.001)and unplanned impulsivity(t=4.960,P<0.001)were significant;the total score in childhood trauma(t=6.603,P<0.001)and the five subscales scores of emotional abuse(t=6.321,P<0.001),physical abuse(t=5.588,P<0.001),sexual abuse(t=5.652,P<0.001),emotional neglect(t=4.021,P<0.001)and physical neglect(t=5.128,P<0.001)were significant differences.4.Emotional neglect,motor impulsivity,and idea of suicide enter the regression equation,and the final model explains about 30%of ASPD symptoms.5.There were statistically significant differences in the genotype frequencies of NOS1AP gene rs945713(?~2=9.577,P=0.008)and allele frequencies(?~2=9.161,P=0.002).6.significant Differences are found in emotional neglect(F=4.340,P=0.014),physical neglect(F=4.989,P=0.007)and total childhood abuse(F=3.635,P=0.028)in different genotypes of NOS1AP gene rs945713.Post hoc testing showed significant differences in emotional neglect and physical neglect between CC and CT/TT,and there was a significant difference in the total score of childhood abuse between CC and TT.7.The genotype of rs945713 mediates between CTQ total score(R~2=0.183,t=-2.294,P=0.023),physical abuse(R~2=0.151,t=-2.510,P=0.013),emotional neglect(R~2=0.115,t=-2.061,P=0.040),physical neglect(R~2=0.151,t=-2.510,P=0.013),and antisocial behavior.Conclusion:1.Male violent criminals with ASPD often violate the law and violate the rules and serve the sentence,smoking and drug use ratio is higher;there are terrible parenting styles;impulsive level is higher,more likely to appear depression,anxiety and suicide.2.Childhood trauma experience will increase the risk of ASPD,in which emotional neglect and motor impulsivity and idea of suicidal have certain predictability for ASPD.3.The CC genotype of rs194713 of NOS1AP gene increases the risk of ASPD,while individuals carrying the CC genotype may be more susceptible to childhood trauma.The interaction between CC genotype and childhood trauma increases the susceptibility of ASPD.
