Objective:To observe the effects of Chaihu Shugan San on the expression of TH,c-fos,AP-1,GR,MR and ER-?in the locus coeruleus,investigate the mechanism of liver stagnation pattern in perimenopausal rats.Method:The peri-menopausal rat model was established by natural aging method and randomly divided into blank group,model group and treatment group.HE staining was used to observe the vaginal exfoliated cells in rats.After the peri-menopausal model was established,the model group and the treatment group were chronically restrained for 42 days,to establish the model of peri-menopausal liver stagnation pattern.From the 21st day,the treatment group used Chaihu Shugan San synchronously.In the intervention,the blank group and the model group were given the same amount of normal saline,and were intragastrically administered once a day for 21 days.The general condition,body weight,behavior and 1%sucrose water preference of rats were observed.The changes of serum CRH,ACTH,CORT,E2,DA and EPI were detected by ELISA.The expression of TH,c-fos,AP-1,GR,MR and ER-?mRNA in rat locus coeruleus was detected by q-PCR.Result:?1?The vaginal exfoliated cells in the pre-emotional rat were mainly nucleated epithelial cells.The vaginal exfoliated cells of estrus rats are mainly non-nucleated keratinocytes.Leukocytes,nucleated epithelial cells and non-nucleated keratinocytes exist in the late estrus.Emotional interval is dominated by white blood cells.he estrous cycle disorder occurred in 12-month-old female rats,suggesting that 12-month-old rats have entered the peri-menopausal period.?2?There was no significant difference in weight,behavior and 1%sucrose water preference between the groups before model establishment.After modeling,the weight of the rats decreased significantly with the increase of modeling time?P<0.05?.Vertical activities,horizontal activities,central residence time and modification activities were all reduced to a certain extent,among which vertical activities,horizontal activities and central residence time were significantly lower?P<0.05?,and 1%sucrose water preference was significantly lower?P<0.01?.After giving Chaihu Shugan San,compared with the model group,the weight loss of the treatment group was slowed down?P>0.05?;the vertical activity of rats and the preference of 1%sucrose water increased significantly?P<0.05?.?3?Serum ACTH and CORT levels were significantly higher in the model group than in the blank group?P<0.05?,and significantly lower in the treatment group than in the model group?P<0.05?.There was no significant difference in serum CRH content between the groups?P>0.05?.Serum E2,DA and EPI levels were significantly lower in the model group than in the blank group?P<0.05?.The treatment group was significantly higher than the model group E2 and DA?P<0.05?,and E2 was increased?P>0.05?.?4?The expression of TH and c-fos mRNA in the locus coeruleus was significantly lower in the model group than in the blank group?P<0.05?.The TH mRNA in the treatment group was significantly higher than that in the model group?P<0.05?,and the c-fos mRNA was increased.?P>0.05?;there was no significant difference in the expression of AP-1 mRNA in each group?P>0.05?;the expression of GR,MR and ER-?mRNA in locus coeruleus was significantly lower in the model group than in the blank group?P<0.05?.Compared with the model group,GR and ER-?mRNA were significantly increased?P<0.05?,and MR mRNA was increased?P>0.05?.Conclusion:?1?Peri-menopausal liver stagnation syndrome rats showed behavioral activity decreased,serum E2,DA and EPI levels decreased,locus coeruleus TH,c-fos,AP-1 and GR,MR,ER-?mRNA expression decreased.It is suggested that the decline of locus coeruleus function plays an important regulatory role in the formation of liver stagnation pattern in perimenopausal rats.?2?Chaihu Shugan San enhances the secretion of catecholamine neurotransmitters by promoting the expression of TH,c-fos and AP-1 in locus coeruleus;promoting the expression of GR,MR and ER-?enhances the negative feedback regulation of HPA axis to jointly enhance the behavioral activities of rats with peri-menopausal liver stagnation syndrome and improve the state of depression.rom the perspective of prescription,it was confirmed that the function of locus coeruleus is closely related to the formation of liver stagnation pattern in perimenopausal rats.
