Objective:The mouse model of allergic rhinitis was established and the miRNA-375 lentivirus overexpression vector was administered during its excitation stage to detect the differential expression of Thymic stromal lymphopoietin(TSLP)in nasal mucosa from the perspective of gene and protein To investigate the effect of miR-375 overexpression to the expression of TSLP in allergic rhinitis and its possible regulatory mechanism.Methods:The ovalbumin(OVA)was used to induced in the allergic rhinitis mouse model and A saline was used in control.MiR-375 overexpression lentiviral vector and control vector were constructed on the basis of induction of mouse allergic rhinitis,and the tail vein injection was performed.The total RNA and total protein were isolated from the nasal mucosa of mice respectively.The mRNA expression of TSLP in the nasal mucosa of each group was detected by real-time fluorescence quantitative PCR with β-actin as a reference gene.Western blotting was performed using GAPDH as a reference protein to detect the relative expression of TSLP in nasal mucosa of each group.Results:Allergic rhinitis mice sensitized with OVA showed symptoms of sneezing,flexing nose and runny nose to varying extents.Histological pathology findings that the nasal mucosa was destroyed and infiltration of inflammatory cells was obvious.After miR-375 overexpression lentiviral vector The above performance is reduced.Real-time fluorescence quantitative PCR showed that the relative expression level of TSLP mRNA in nasal mucosa of miR-375 lentivirus overexpression group was significantly lower than that of allergic rhinitis group and control vector group(P<0.05).The relative expression levels of TSLP mRNA in miR-375 lentiviral overexpression group,control vector group and allergic rhinitis group were significantly higher than those in saline control group(P <0.05).Western blotting showed that TSLP protein expression in nasal mucosa of allergic rhinitis group was significantly higher than that of saline control group(P <0.05),miR-375 lentivirus overexpression vector significantly decreased(P <0.05)However,TSLP protein expression was also down-regulated in control vector group compared with that in allergic rhinitis group(P <0.05).The difference was statistically significant.Conclusions:In this study,the relationship between miRNA-375 and TSLP was explored.Overexpression of miR-375 in nasal mucosa of allergic rhinitis mice showed negative regulation of TSLP expression,which may be involved in the regulation of Th2 cell differentiation Immune response,which is provide experimental reference and theoretical basis to TSLP-targeted allergic rhinitis treatment.
