Understanding mechanisms of cancer metastasis is crucial for reduction of cancer mortality.This thesis mainly discusses the function and prognosis of ACSM3 and LASS2 in hepatocellular carcinoma,and briefly introduced the influence of ANGPTL1 on migration and invasion of hepatocellular carcinoma.Aims: The thesis consists of three parts.The first part investigated the expression and prognosis value of ACSM3 in HCC,and also addressed underlying roles of ACSM3 in HCC and molecular mechanisms of its biological functions.The second part analyzed the value of combined detection of LASS2 and TGF-?1 in HCC prognosis.The third part investigated the expression and potential roles of ANGPTL1 in HCC.Methods: In first part,expression of ACSM3 was investigated by real-time PCR and western blot.Kaplan-Meier analysis and Cox's proportional hazards regression model were used to calculate the effects of ACSM3 on HCC prognosis.Migration and invasion assays were carried out for functional analysis in vitro and a xenograft model was used to analyze the effects of ACSM3 on cancer metastasis in vivo.Furthermore,human phospho-kinase array assays were performed to explore molecular mechanisms of ACSM3 in HCC.In second part,Kaplan-Meier analysis and Cox's proportional hazards regression model were applied to analyze the value of combined detection of LASS2 and TGF-?1 in HCC prognosis.In third part,expression of ANGPTL1 was investigated by real-time PCR and western blot.Migration and invasion assays were carried out for functional analysis on cell motility in vitro.Results: ACSM3 expression in tumorous tissue was obviously downregulated compared to adjacent non-tumorous liver tissue of HCC patient,and ACSM3 was an independent prognostic factor for HCC.ACSM3 suppressed metastasis of HCC by AKT-WNK1 signaling pathway.Combination of LASS2 and TGF-?1 was an independent prognostic marker for HCC.ANGPTL1 was obviously downregulated in tumorous tissue compared to adjacent non-tumorous liver tissue of HCC patient.Overexpression of ANGPTL1 suppressed migration and invasion of HCC cells in vitro.Conclusion: This study indicates that ACSM3,LASS2 and AN GPTL1 served as tumor suppressor molecules play an important role in HCC.
