Objective:To detect the expression of methylation transferase 3A(DNMT3A)in Gastric cancer(GC)tissue,adjacent tissues,chronic non-atrophic gastritis(CNAG)tissue and chronic atrophic gastritis(CAG)in Han and Tibetan,and to analyze the difference of DNMT3 A expression between Chinese and Tibetan communities,and to explore the mechanism of DNMT3 A involved in the occurrence of gastric cancer.Methods:30 cases of Han and Tibetan GC patients diagnosed by gastroscopy and pathological biopsy were selected as the control group in 30 cases and 30 cases of gastric mucosa in CNAG and CAG patients,and real-time fluorescence quantitative PCR was used to detect DNMT3 A expression in the experimental group and control group.Results: 1.The relative expressions of DNMT3 A in GC group,adjacent tissue group,CNAG Group and CAG Group were: 1.91(1.12,3.26),0.70(0.52,1.16),0.69(0.50,1.01),0.87(0.56,1.75),respectively.The relative expression of DNMT3 A in GC tissue was significantly higher than that in the adjacent tissue group,CNAG Group and CAG Group(p<0.05),and there was no statistically significant difference between the relative expression of DNMT3 A in the adjacent tissue group,CNAG Group and CAG Group(p>0.05).2.The relative expression of DNMT3 A in gastric mucosal biopsy of Han GC group,adjacent tissue group,CNAG Group and CAG Group was 1.97(1.09,3.31),0.73(0.52,1.47),respectively.0.71(0.53,0.97),1.00(0.50,1.71),the relative expression of Tibetan lesions in the same level was 1.91(1.20,3.25),0.66(0.54,1.14),0.60(0.49,1.81),0.87(0.60,1.99).There was no statistical difference in the expression of DNMT3 A in gastric mucosal biopsy in two ethnic groups(P>0.05).3.The expression of DNMT3 A in GC tissue was related to the differentiation degree of GC,lymph node metastasis(p<0.05),but not to the patient's sex,age,tumor size,infiltration depth,distant metastasis,TNM staging,and the difference was not statistically significant(p>0.05).The expression of DNMT3 A in low differentiation GC tissue was significantly higher than that in middle and high differentiation GC(p<0.05),and the expression of DNMT3 A in lymph node metastasis GC was higher than that of GC tissue(p<0.05)without metastasis of lymph nodes.Conclusions: 1.There was no significant difference in the expression of DNMT3 A in the same grade of gastric mucosal biopsy in Han and Tibetan,suggesting that ethnic factors might not affect the expression of DNMT3 A.2.The expression of DNMT3 A was not different in chronic non-atrophic gastritis,chronic atrophic gastritis and adjacent tissues,and was higher in gastric cancer tissue than chronic non-atrophic gastritis,chronic atrophic gastritis and adjacent tissues,indicating that DNMT3 A was involved in the development of gastric cancer.3.The expression of DNMT3 A in low-differentiation GC tissue was significantly higher than that in middle and high differentiation GC,and DNMT3 A expression in lymph node metastasis GC was higher than that of lymph node without metastasis,suggesting that DNMT3 A was involved in the progression and migration of gastric cancer.Dynamic monitoring of the expression level of DNMT3 A may provide some reference for judging the progression stage and prognosis of gastric cancer.
