Objective To investigate the effect and mechanism of inulin on alcoholic liver disease in mice.Methods Female C57BL/6J mice were randomly divided into 4 groups(n=15/per group): negative control group,model group,positive control group and intervention group.After 6 weeks of feeding,mice were euthanized and associated indications were researched.The protective effect of inulin on liver injury induced by alcohol was evaluated by measuring the final body weight of mice,detecting plasma alanine aminotransferase(ALT)and aspartate aminotransferase(AST)levels,and observing HE staining of liver paraffin sections.Limulus amebocyte lysate test and flow cytometry were used to measure the levels of plasma lipopolysaccharide(LPS)and the differences of various inflammatory indicators(IL-6,IL-17 A,TNF-?,IL-10,M? and TLR4)among diverse groups.The immunohistochemistry was used to detect the levels of liver macrophage(M?)in diverse groups.High-throughput 16 S rRNA sequencing was aimed to detect the diversity and abundance of gut microbiota in mice.Results The final body weight of model group was significantly lower than negative control group or positive control group,but intervention group was higher than model group.The ALT and AST levels in model group was notably higher than negative control group or positive control group,but the transaminases in intervention group was lower than model group.The levels of plasma and liver pro-inflammatory indicators including LPS,TNF-?,IL-6,IL-17 A,M? and TLR4 were also significantly increased in model group.The levels of anti-inflammatory cytokine IL-10 were significantly reduced.However,after the intervention of inulin,the above-mentioned pro-inflammatory indicators were significantly decreased but IL-10 was notably higher than before.HE staining of the liver sections found that inulin can also effectively alleviate steatosis and steatohepatitis in ALD mice.In addition,in the model group,compared with the negative control and the positive control groups,the mouse intestinal Proteobacteria and Actinobacteria were significantly increased at phylum level,the Allobaculum,Lactobacillus and Lactococcus were significantly reduced while Parasutterella was significantly increased at genus level.However,the reduction of Proteobacteria,Actinobacteria and Parasutterella,as well as increase of Allobaculum,Lactobacillus and Lactococcus in the intervention group than that in model group.Conclusion Inulin may alleviate the progression of ALD by suppressing of inflammatory cascade and regulating the gut microbiota.
