Gut microbiota are microorganisms resident in the intestinal tract,which compete and cooperate with each other and come to a balance.On the one hand,gut microbiota composition is impacted by diet,antibiotics,genetic background and external environment.On the other hand,gut microbiota affect host through metabolic activity.The two most important factors build gut microbiota composition are diet and genetic background,which function together to impact gut microbiota and play an important role in host health.Diet dominant genetic background in shaping gut microbiota composition.The mouse gut microbiota composition and metabolomics have significant change in high fat diet condition compared to normal diet condition.The three main short chain fatty acids,acetate,propionate and butyrate concentration decrease,while the branch short chain fatty acid isobutyrate and isovalerate increase in mouse feces.Medium chain and medium-long chain inulin have similar prebiotic signature,while short chain inulin is distinctive.The most distinctive bacteria affected by inulin addition was S247,which was more abundant in tree chain inulin group.Though the polymerization degree lead to difference in gut microbiota composition,the microbiota metabolomics change was similar.10 % short,medium or medium-long chain inulin supplement in high fat diet condition can significant alter the mouse fecal metabolomics and rescue the short chain fatty acid level to relative normal level.More than 200 known compounds in library were measured relatively in fecal metabolomics analysis and more than 100 metabolites were found have significant change and similar trend in tree chain inulin group.Identified metabolites were enriched and assigned to KEGG pathways and matched with 16 S rDNA genome PICRUSt results.The identified pathway concentrated on amino acid metabolism,vitamin metabolism and fatty acid metabolism.Inulin addition didn't cause significant change in body weight,blood glucose and serum TAG level,while basic metabolic rate elevated,fat mass decreased,serum NEFA decreased and carnitine and acylcarnitines level elevate in mouse.
