Objective: To study the effect of LASS2 on proliferation and apoptosis of papillary thyroid carcinoma(PTC)cell.Methods: 1.The expression level of LASS2 in 60 PTC tissues,20 paracancerous tissues and 37 nodular goiter tissues were detected by immunohistochemistry.2.Recombinant adenovirus Adv-LASS2-GFP was constructed and transfected into BCPAP cells as the experimental group.Meanwhile,blank control group(NC)and no-load group(Adv-GFP)were set up.The expression levels of LASS2 mRNA and protein in BCPAP cells of each group were detected by Real-time fluorescence quantitative polymerase chain reaction(RT-qPCR)and Western Blot(WB).3.CCK-8 and flow cytometry were used to detect the proliferation,cell cycle and apoptosis of BCPAP cells in each group,and WB was used to detect the expression of CyclinD1 and CDK4 in each group.Results: 1.There were statistically significant differences in LASS2 protein expression in PTC,paraneoplastic thyroid carcinoma and nodular goiter(P<0.05).2.Overexpressing of LASS2 gene in BCPAP cell was successfully constructed.3.The CCK-8 experiment suggested that overexpression of LASS2 suppressed the proliferation of BCPAP cells(P<0.05).Flow cytometry indicated that G0/G1 phase group and the Adv-GFP group(P<0.05).Flow cytometry indicated that overexpression LASS2 promoted apoptosis(P<0.05).WB detection of cell cyclin-related proteins suggested that: in the group of Adv-LASS2-GFP,the CyclinD1 and CDK4 were lower than those in the NC group and the group of Adv-GFP,and the difference was statistically significant(P<0.05).Conclusion: LASS2 expression was down-regulated in PTC and correlated with clinical stage and cervical lymph node status(P<0.05).Overexpression of LASS2 suppresses BCPAP cell proliferation,promotes apoptosis,and induces G0/G1 cell cycle arrest,which may be related to the down-regulation of CyclinD1 and CDK4.
