ABSTRACTFollowing the clincal experience of Professor Zou Yunxiang and his recipe, Baoshen tablet (BST) is bused on the treatment principle of protecting Kidney Yin and Kidney Yang. The total effective rats is 76. 7%. The former study suggested that BST could inhibit the proliferation of rat mesangiul cells and flbroblasts, and significantly inhibit the expression of transforming growth factor ?(TGF) and type I and type N collagen of extracellular matrix (ECM). Objective: To discuss the effect of BST on mesanginl cell and fibroblast in culture, and reveal its mechanism on the level of cellulomolecule. Methods: Got SD rats kidney for culture of fibroblasts, added the rats serum with different concentration of BST to fibroblasts, added the human serum from healthy volunteers with diHerents concentration of BST to mesangial cells. The proliferation, apoptosis, and cell cycle of mesangial cell and fibroblast were deteeted by MTT assay and flow cytometny respectively. Results ~ BST inhibited the proliferation of mesangial cells and fibroblasts significantly not by cytotoxxn. It transformed the cell cycle, delayed the process of cell cycle, and -inhibited cell proliferation by calcium pathway inhibition to decrease calcium intraflowing. and decreasing the signal transduction amony regulating factorsin cell cycle. BST could induce fibroblast and mesangial cell apoptosis, and could reduce the cell aggregation due to enhanced mitogen, reduce the number of fibroblasts and mesangial cells by cell apoptosis. Conclusion: BST has significant effect on inhibiting proliferation of flbroblasts and mesangial cells and pomotixig apoptosis of them. Its inhibiting the activity of TGF and reducing the growth of ECM may be one of the mechanism of its clinical curative effect. Longterm administration of BST may be benificial to glomerulosclerosis and renal inlerstitial fibrosis in chronic renal failure (CRF), so that to prevent and reduce the renal scar tissue formation and returd the occurrence of end stage CRF.
