Breast cancer is one of the most prevalent malignant tumors in global females,and its mortality is increasing year by year,besides that we found the age of onset is yonger.The 5 years survival rate of breast cancer is higher than other cancers in clinical,such as lung cancer,stomach cancer,colorectal cancer and so on.At present,there are 4major treatments,for example:surgery,radiotherapy,chemotherapy and biotherapy.Chemotherapy plays an indispensable role in the treatment of breast cancer,both preoper-ative neoadjuvant and postoperative adjuvant chemotherapy are very important.However,in clinical,with the increase of chemotherapeutic dose,the side effect also become more and more stronger,which will only bring physical pain,but also increase the psychological burden to patients.So we should research a new kind of drug with low toxicity and high efficiency,but the development of new drugs is a long process,so the scheme of drug combination is also necessary.Aspirin is a kind of NSAIDs,in the beginning it was used for antipyretic analgesia,meanwhile it is also the basic drug of anti-thrombus in cardiovascular disease;recently a number of researches indicated that NSAIDs not only could prevent the occurrence of tumors,but also suppress the growth of tumor and promote the apoptosis of tumor cells.The effects of anti-tumor had reported in lung cancer?stomach cancer?liver cancer?colorectal cancer?breast cancer and so on.Objective:To explore the application of non selective cyclooxygenase inhibitor in breast cancer and its exact antitumor effect by the study of non steroidal anti-inflammatory drug(aspirin)in human breast cancer MCF-7 cells,and provide more reliable basis for the application of aspirin in breast cancer.Methods:we divided the experiments into control group,experimental control group,monotherapy cyclophosphamide group,monotherapy aspirin group,aspirin combines cyclophosphamide group.Then we observed and measured the anti-tumor effect of aspirin and cyclophosphamide on MCF-7 breast cancer cells by CCK-8 assay.In addition,we al-so detected the expression of p53 and NF-k B on MCF-7 breast cancer cells by the assay of Western-Blot and Immunofluorescence.Results:1.The anti-tumor effect of aspirin and cyclophosphamide on breast cancer MCF-7 cells by CCK-8 assay.(1)Aspirin could inhibit the growth of breast cells MCF-7,which was positively correl ated with time and dose(p<0.05).Low-dose(0.5-2mmol/L)aspirin had no effect on br-east cancer MCF-7 cells,but as the dose increased,the inhibition effect of breast cancer MCF-7 cells was more and more significantly,and this effect was positively correlated with time and dose.(2)Cyclophosphamide could inhibit the growth of breast cancer MCF-7 cells,which w-as positively correlated with time and dose(p<0.01).(3)The results of different concentrations of cyclophosphamide and aspirin on MCF-7cells 48 h respectively:the IC50 of cyclophosphamide was 2umol/m L,and the IC50 of aspirin was 2-5ummol/L(p<0.01).(4)The effect of low concentration aspirin(0.5mmol/L)combined with cyclophosphamide(2ummol/m L)on MCF-7 breast cancer cells was higher than the effect of aspirin or cyclophosphamide respectively(p<0.01).2.The assay of Western-Blot and Immunofluorescence detected the expression of p53 and NF-?B on breast cancer MCF-7 cells(1)The expression of p53 was up-regulated,but NF-?B was down-regulated on MCF-7 breast cancer cells after treatment with low concentration of aspirin(p<0.05).(2)The expression of p53 was up-regulated,NF-?B,however,was down-regulated on MCF-7 breast cancer cells after treatment with cyclophosphamide(p<0.01).(3)The expression of p53 was up-regulated,while NF-?B was down-regulated on breast cancer MCF-7cells after treatment with aspirin combined with cyclophosphamide(p<0.01).Conclusions:Aspirin had a synergistic effect with cyclophosphamide,which not only inhibited the growth of tumor cells,but also promoted their apoptosis.
