Objective: To explore the change of the expression level of PAPPA2 in pregnancy in patients with severe pre-eclampsia and its effect on the proliferation,invasion,migration and network formation of extravillous trophoblasts.Methods:(1)In September 2016 to October 2017 in Huazhong University of Science and Technology Tongji medical school affiliated Tongji hospital of Obstetrics and Gynecology childbirth 34 weeks to 40 weeks of gestation with severe PE and gestational age of control group placental tissue,and were applied Western bloting,Real-time PCR and immunostaining techniques to detect PAPPA2 m RNA and protein expression level.(2)Using HTR-8/SVNEO cell lines,immunohistochemistry experiments were performed to detect the expression of PAPPA2 in extravillous trophoblasts,and to analyze the effects of PAPPA2 on the proliferation,invasion,migration and network formation of HTR-8/SVNEO cell lines.(3)The expression of PAPPA2 protein was down-regulated by Si RNA,and the changes of HTR-8/SVNEO cell invasion,migration and network formation were observed.Results:(1)The PAPPA2 m RNA levels of the normal control group and the case group were respectively0.723±0.085and1.545±0.045,P<0.05;The PAPPA2 protein expression levels were respectively1.528±0.342and2.983±0.376,P<0.05.PAPPA2 protein is mainly expressed in placental trophoblasts.(2)The PAPPA2 protein was expressed in the HTR-8/SVNEO cell lines,and the main expression was cytoplasm and membrane.(3)The PAPPA2 protein has no obvious effect on the proliferation of HTR-8/SVNEO cell lines,and it has inhibitory effect on migration,network formation and invasion.(4)When the si RNA was used to down-regulate PAPPA2,the migration,invasion of the HTR-8/SVNEO cell line were enhanced,but the proliferation was not statistically significant(P >0.05)compared with the control group.Conclusion: PAPPA2 m RNA and protein expression in placental tissues of patients with severe preeclampsia were increased.In vitro experiments,PAPPA2 protein inhibits the migration,invasion,and network formation of extravillous trophoblasts..
