Abstract/Summary:
ObjectiveAbnormal lipid metabolism,the mainly clinical manifestation is hyperlipidemia,is the starting point of cardiovascular diseases(CVD)such as atherosclerosis,and is a major component of metabolic syndrome.The occurrence of hyperlipidemia is closely related to nutritional factors.Both selenium and riboflavin play positive roles on hyperlipidemia by improving levels of lipid metabolism regulators and lipid metabolism enzymes.Previous reports have focused on the relationship between single nutrients and hyperlipidemia.There are few reports about the effects of combined intervention of two or more nutritional factors on hyperlipidemia.This study is to investigate the effects of sodium selenite combined with vitamin B2 on lipid metabolism in hyperlipidemia rats,providing a scientific basis for the treatment of hyperlipidemia and providing evidence for the joint application of nutrients co-treatment on hyperlipidemia.MethodsA total of 50 male Sprague Dawley(SD)hyperlipidemia model rats were divided into five groups,including model group(HF),low-dose sodium selenite and low-dose riboflavin combined intervention group(LSe+LVB2),high-dose sodium selenite and low-dose riboflavin combined intervention group(HSe+LVB2),low-dose sodium selenite and high-level riboflavin combined intervention group(LSe+HVB2)and low-dose sodium selenite and high-dose riboflavin combined intervention group(HSe+HVB2).The dose of LSe,HSe,LVB2 and HVB2 were 0.83 μg/kg BW?d,8.33 μg/kg BW?d,0.70 mg/kg BW?d and 3.50 mg/kg BW?d.10 male rats in normal blood lipid were divided into wild type group(WT).All rats were treated by gavage for 60 days.Wild type group and model group were given saline.The serum of rats was collected at the 20 th,40th,and 60 th day of intervention for determination of blood lipid,and the Apo A1,Apo B,ALT and AST were determined at the end of treatment.The liver homogenate was made for measurements of liver lipid and the liver tissue was stained by HE and Oil Red O for pathological examination..The activities of hepaticlipase(HL)and lipoprotein lipase(LPL)in serum and the level of 3-hydroxy-3-methyl glutaryl coenzyme A reductase(HMGR)in liver homogenate were determined at the end of treatment.The m RNA expression levels of hepatic enzymes(ACAT,LCAT,LDLR,PPARγ and SREBP-1c)and the levels of protein expression including SIRT1,CAV,CEL,PPARγ and SREBP-1c were measured by q PCR and Western blot,respectively.ResultsAt the end of the modeling period,the levels of serum TC,TG,LDL-C,ALT and AST and the levels of liver tissue TC,TG,LDL-C in the model control group were significantly higher than the wild type rats.Pathological examination showed the degree of liver steatosis and hepatic intracellular triacylglycerol accumulation in model group was higher than the wild rats.The result showed that the rats of model group were in hyperlipidemia which was suitable for later study.The result of blood lipid and apolipoprotein showed that all of ways of intervention could decrease the level of blood lipid,promote secretion of Apo A1,inhibit secretion of Apo B and increase the ratio of Apo A1/Apo B.Compared with other intervention groups,the improvement in LSe+LVB2 intervention group for lipid metabolism was more significant.The results of esterase in serum and lipid metabolism enzyme in liver tissue showed the level of HMGR in liver homogenate was declined and the level of serum HL was increased of hyperlipidemia rats though combined intervention of sodium selenite and Vitamin B2.Sodium selenite and riboflavin can regulate the m RNA expression level of lipid metabolism factor,which promotes the m RNA expression level of ACAT and inhibits the m RNA expression levels of HMGR,SREBP-1c and PPARγ.Compared with the model group,combined intervention of sodium selenite and riboflavin can inhibit the protein expression levels of PPARγ and SREBP-1c in the liver.Compared with other intervention groups,the regulation in LSe+LVB2 intervention group for lipid metabolism factors were more significant.Conclusion1.The combined intervention of trace element selenium and vitamin B2 can effectively reduce the level of blood lipid and regulate lipid metabolism in hyperlipidemic rats,which has a certain protective effect on non-alcoholic liver disease caused by long-term high-fat diet.2.The possible mechanism of the combined intervention of selenium and vitamin B2 in reducing blood lipid in hyperlipidemic rats is to regulate the metabolism of cholesterol,triglyceride and fatty acid by inhibiting the expression of PPARγ and SREBP-1c,the key lipid metabolic regulators,and affecting the activities of lipid metabolic enzymes such as HMGR and HL.
