Evaluation Of Preventing Effect Of Sanhuang Shu'ai Decoction In DSS Induced Ulcerative Colitis Mice

Aim:To evaluate the preventive effects of San huang Shu'ai Decoction on ulcerative colitis at the whole and molecular level using the mice ulcerative colitis?UC?model which was induced by dextran sodium sulfate?DSS?.To provide a theoretical basis for the development of Sanhuang Shu'ai Decoction as a ulcerative colitis medicine in the future.Method:Establishing the mice model of ulcerative colitis induced by DSS and evaluate the role of Sanhuang Shu'ai Decoction in ulcerative colitis according to disease activity index,colon length and histopathological score.Elisa,Western Blot and immunohistochemistry were used to detect the expression of inflammatory factors and related cell signaling pathways in colon tissues.The contents of MDA,MPO and hydrogen peroxide in colon tissues were measured by physical and chemical methods.At the 8^th day,the mice's feces were collected and frozen for 16S rDNA sequencing and analysis.The inflammatory cell model was established to analysis the effect of Sanhuang Shuai Decoction on ROS production by the fluorescence probe method.Result:The DAI and colon pathological condition of mice treated with Sanhuang Shu'ai Decoction were significantly slighter than those of model group.Sanhuang Shu'ai Decoction significantly reduced the contents of IL-1?,IL-6 and TNF-?in colon tissue of the mice,inhibited the phosphorylation and nucleation of NF-?b,and decreased the levels of MPO and MDA.Sanhuang Shuai Decoction has obvious protective effect on intestinal flora abundance and lactobacillus,and can also increase the levels of hydrogen peroxide in tissues and ROS in cells.Conclusion:Sanhuang Shu'ai Decoction can significantly alleviate ulcerative colitis induced by DSS.Its mechanism may be related to inhibiting oxidative stress level,inhibiting NF-kb signaling pathway,protecting intestinal flora and increasing hydrogen peroxide level.
