| ã€Objective】By means of ulcerative colitis(UC) patients and healthy human gut macrobiotic diversity, similarity and species composition and so on; we confirmed deficiency and excess syndromes of UC in Traditional Chinese medicine(TCM) have different structures in intestinal microflora. It provided the material basic pathological mechanism for the excess and deficiency syndrome of UC theoretical and experimental through analyzed correlation between excess and deficiency syndromes and their differences structure of intestinal flora,ã€Methods】We use UC patients and healthy volunteers as the research object, and established of TCM syndrome diagnosis, inclusion and exclusion criteria, divided them into spleen-stomach deficiency syndrome group, internal retention of dampness-heat syndrome group, spleen deficiency and dampness-heat syndrome group and the healthy control group, then the total DNA was extracted from stool collection; 16 Sr DNA V3 PCR amplification, denaturing gradient gel electrophoresis(DGGE analysis of intestinal flora) DNA fingerprint; Mi Seq sequencing technology to carry out the intestinal flora of 16SV3+V4 region sequencing and bioinformatics analysis of DNA fingerprinting and 16SV3+V4; bioinformatics data based on the analysis of different syndrome UC patients and healthy population of the structure of intestinal microflora and correlation.ã€Results】The DNA fingerprint analysis of intestinal flora showed that the UC patients and the health of human intestinal flora structure have significant difference, UC patients with intestinal flora diversity and abundance were higher than those in healthy group(P < 0.05), the intestinal similarity of healthy group is higher than that of UC group(P < 0.05), the diversity and similarity between three UC groups and control group had significant difference(P < 0.05), Firmicutes, Bacteroides and Proteobacteria three dominant bacteria composition ratio have significant difference between the four groups(P < 0.05).16SV3+V4 sequencing and bioinformatics analysis showed that: the Bacteroidetes and Firmicutes proportion of UC patients relative to other groups was large quantity constant basically, but there were significant differences in their accounts for different excess and deficiency syndrome group. Firmicutes intrinsic proportion in internal retention of dampness-heat syndrome group is lower than the healthy group(P < 0.05), Veillonella which is one kind of species of Firmicutes, it is similar with Firmicutes(P < 0.05), but it in the spleen-stomach deficiency syndrome group contains a higher proportion than internal retention of dampness-heat syndrome group(P < 0.05). The proportion of Proteobacteria in spleen deficiency-dampness heat group is higher than the spleen deficiency group(P < 0.05). The proportion of Haemophilus influenzae Bacillus in the internal retention of dampness-heat syndrome group is higher than the spleen deficiency group and spleen deficiency and dampness heat group(P < 0.05). Bacteroidetes, Bacteroides and Prevotella accounted for in the four groups have different proportions. Bactericides accounted for in damp heat syndrome group was higher than that healthy control group(P < 0.05). The proportion of Prevotella subjects in all ulcerative colitis accounted for intestinal flora were lower than those of healthy controls(P < 0.05). Put the Corynebacterium as an example, it is a specific flora of actinomycetes,it in the internal retention of dampness-heat syndrome group higher than the proportion of spleen qi deficiency group(P < 0.05). Bacteria accounted for in the spleen and stomach deficiency group and spleen deficiency-dampness heat group was less than the proportion of healthy group(P < 0.05).DGGE and the sequencing results similarly reflect the structure of intestinal flora of UC patients with different excess and deficiency syndrome group and healthy group.ã€Conclusion】1. Comparison of three kinds of syndromes of UC patients and healthy people, the structure of intestinal flora, such as similarity and diversity are differences.2. Three kinds of deficiency and excess syndromes of UC patients and healthy people, their composition of intestinal flora from Phylum level to species level of bacterial are different, may be because the proportion of Bacteroidetes and Firmicutes and prevotella in intestinal had significant difference.3. Compare three kinds of syndromes of UC patients, the composition of intestinal flora at phylum level of bacterial are different because of the proportion of Firmicutes and Proteobacteria in intestinal had significant different. |
