Objective:To investigate the changes of biochemical indexes and blood gas analysis in obese pre-type 2 diabetes mellitus(T2DM)insulin resistant(IR)rats and Gegen Qinlian decoction(GQD)treated with high fat diet.To clarify the therapeutic effect of GQD on IR and obese T2 DM in a timely manner.Methods:Male SD rats were fed with 36% fat-fed high-fat diet for 12 weeks to induce the obese IR mode.After fasting for 12 hours,blood samples were collected from the posterior venous plexus and serum was separated to detect biochemical indexes.After anesthesia with 10% chloral hydrate,blood samples were collected from the posterior venous plexus and the serum was separated,and 10% chloral hydrate was anesthetized.The abdominal aorta was taken for blood gas analysis.The male SD rats were randomly divided into two groups: model group and drug-treated group after they were fed with 60% fat-fed high-fat diet for 12 weeks when the obese pre-T2 DM model with IR was induced?GQD was given for 16 weeks,and the rats were fed with GQD for 16 weeks.At the same time,the model group was fed with high fat diet to form obese T2 DM.After fasting for 12 hours every 4 weeks,blood samples were collected from the venous plexus and serum was separated,biochemical indexes were detected.After 16 weeks of administration,10% chloral hydrate was used to anesthetize the abdominal aorta for blood gas analysis.Results:In obese IR rats,there was no significant difference in pO2,pCO2,sO2 between IR group and normal group(P > 0.05).Hct,ctHb,ctO2 in IR group was significantly higher than that in normal group(P < 0.05).IR).There was a significant positive correlation between Hct,ctHb and HDL-C in obese pre-T2 DM IR group.After the rats with obese pre-T2 DM IR were fed with high fat diet for 24 weeks,the FPG of the model group was increased(P < 0.05),and the obese T2 DM was formed.After 16 weeks of administration,compared with the model group,GQD could control the body weight of rats fed with high fat diet(P < 0.05),when middle dose and high dose were effective fast,middle dose and low dose lasting and stable;low dose of GQD control the blood glucose level of obese T2 DM rats(P < 0.05);.Low and middle dose of GQD could improve the IR(P level of obese rats(P < 0.01).GQD could reduce TC(P(P < 0.01),LDL-C(P < 0.01)and HDL-C(P < 0.01).In obese T2 DM rats,the blood pO2,Hct,ctHb,ctO2 of abdominal aorta was increased(P < 0.01),but there was no significant difference in sO2(P > 0.05).Low dose of GQD increased pH and sO2 levels,(P < 0.05).In the model group,Hct,ctHb,ctO2 was negatively correlated with FINS and pO2 was negatively correlated with TC,HDL-C,LDL-C.Conclusion:in obese IR rats,there is no hypoxia in circulation,but the level of Hct,ctHb,ctO2 is increased.There is a positive correlation between Hct,ctHb,ctO2 and HDL-C.GQD can effectively control the weight gain caused by its high fat diet and delay the development of the early course of obese T2 DM.However,if GQD is used to intervene the obese pre-T2 DM,attention should be paid to the dosage and the duration of administration of GQD.The low dose of GQD increased sO2 in obese T2 DM rats,suggesting that internal dampness in the course of development of obese T2 DM may be related to lung oxygenation and oxygen carrying function of red blood cells.Obese T2 DM rats had no hypoxia in circulation and increased Hct,ctHb,ctO2,which was consistent with obese insulin resistance rats,and increased pO2 at the same time.In obese T2 DM rats,Hct,ctHb,ctO2 was negatively correlated with FINS and pO2 was negatively correlated with TC,HDL-C,LDL-C.
