Breast cancer has become one of the most common malignant tumors that threaten the health of women worldwide.The investigation of molecular mechanisms underlying the breast cancer progression is critical for the treatment of breast cancer.As a member of the IPP complex,PINCH1 protein participates in many signaling pathways in cells and plays an important role in cell growth,migration and proliferation.Our study found that PINCH1 protein is highly expressed in breast tumor tissues and plays an important role in the development of breast cancer.By generating a PINCH1 breast tumor-specific knockout mouse model,we found that knockout of PINCH1 has an inhibitory effect on the development and metastasis of PyMT oncogene-induced breast cancer.We found a novel protein Myoferlin that can interact with PINCH1 in the MDA-MB-231 human breast cancer cell line by IP/MS method and found that the expression of Myoferlin is regulated by PINCH1.Myoferlin is also one of the key molecules in the evolution of breast cancer.PINCH1 is likely to affect the development and progression of tumor by regulating Myoferlin signaling pathway.However,the specific mechanism remains to be further studied.In general,this research is the first in vivo study to demonstrate that PINCH1 plays an important role in the development of breast tumors.And the finding of new binding protein of PINCH1?Myoferlin provides a new clue for the mechanism underlying PINCH1 regulation of the breast cancer.
