Abstract/Summary:
Extracorporeal Shock Wave Therapy(ESWT)is an noninvasive therapy and has been used to fracture nonunion treatment for several decades.In matter of fundamental research,scholars have some knowledge of ESWT’s osteogenesis and osteoclast effects while knowing little about its bone remolding effects.Our research put MC3T3-E1 and RAW264.7 together on the substitution of bone to create a co-culture system.We use ESW to intervene on this system and then evaluating the system’s proliferation via CCK8 test.Scanning electron microscope and fluorescent confocal microscope were used to observe cells’ morphology.PCR was used to detect gene expression of the coculture system.The result show that ESWT activates RAW264.7 in early stage and promotes MC3T3-E1’s proliferation and differentiation.Therefore,we can conclude that one way of ESWT cure nonunion is though bone remolding.In matter of clinical research,ESWT is individual.Some patient didn’t response to ESWT and missing the best procedure time.We observe nonunion(including gap of nonunion,callus hardness,local blood supply and hematoma formation)via ultrasound before ESWT and formatting a system for prognosis valuating via statistical analysis: healing grade=-0.197×gap of nonunion+0.328×local blood supply-0.42×callus hardness/ESWT energy intensity+ 0.956×hematoma formation+3.053...
