Objective To observe the effect of propofol on the expression of PSD-95 in spinal cord segment of L4-6-6 in mice with sciatic nerve detachment injury.Methods 144 cleansing-grade healthy male BALB/c mice were selected,the body weight was20±2g with 8 norfloxacin,and the mice were divided into 3 groups by random Digital scale method:Sham Operation Group?group S?,model group?group M?,propofol Group?group P?,48 in each group,obsession time was 1W,2W,4W,8W,respectively.Each point in time is 4,and a classical sciatic nerve injury model was established by end-to-end anastomosis after breaking the right sciatic nerve.In group P,the peritoneal injection of propofol 50mg·kg-1·d-1was carried,group M was injected with same volume saline after the successful mold,and the continuous intervention was 7 days.The expression of PSD-95 mRNA and protein in L4-6spinal cord segment of mice was detected by real-time PCR method and Western Blot method,and the morphological changes of sciatic nerve myelin sheath in mice were observed by LFB staining.The neurons in the spinal cord segment of L4-6-6 were labeled by TUNEL staining and the apoptosis of neurons was observed,and the wet quality of gastrocnemius muscle in the right and left of each mouse was reported respectively,and the residual rate of wet mass of gastrocnemius muscle was calculated.Results 1.PSD-95 mRNA results showed that compared with group S,the expression of PSD-95 mRNA in group M was up-regulated at 1W,2W and 4W?P<0.05?.Compared with group M,the expression of PSD-95 mRNA in group P was significantly down-regulated at1W,2W and 4W?P<0.05?.2.The results of PSD-95 Western Blot showed that the expression of PSD-95 protein in group M and group P was up-regulated at 1W;the expression of PSD-95protein in group M reached the peak at 2W;compared with group S,2W The expression of PSD-95 in group M was significantly up-regulated with 4W?P<0.05?.Compared with group M,the expression of PSD-95 was down-regulated at 2W and 4W?P<0.05?3.LFB staining showed:compared with group S,the sciatic nerve myelin sheath in the injured section of group M was messy,the thickness of myelin sheath was uneven and the contour was incomplete,and compared with the group M,the morphology of myelin sheath and thickness contour of group P were improved.4.The apoptosis of L4-6-6 segment neurons in the spinal cord of mice by TUNEL staining showed that with the prolongation of postoperative time,The number of neuronal apoptosis in the L4-6-6 segment of spinal cord decreased gradually?P<0.05?,and compared with group S,the number of apoptosis of spinal cord neurons in Group m and group P increased significantly?P<0.05?,and the number of apoptosis of nerve cells in the L4-6-6 segment of spinal cord in group P decreased significantly?P<0.05?compared with group M.5.The residual rate of wet quality of gastrocnemius muscle showed that compared with group S,the residual rate of wet mass of gastrocnemius muscle in group P was decreased?P<0.05?at 2W,4W and 8W,and the residual rate of wet mass of gastrocnemius in group P at 2W,4W and 8W was increased?P<0.05?compared with group M.Conclusion1.The possible mechanism of propofol promoting neurological recovery is through inhibiting the expression of PSD-95.2.Propofol can reduce the apoptosis of corresponding spinal cord neurons after sciatic nerve injury...
