Objective To evaluate the positive expression rate of PTEN(Phosphatase and tensin homolog),MTDH(Metadherin)and GATA3(GATA binding protein 3)in normal colorectal samples,colorectal adenoma samples and colorectal cancer samples.To explore the positive expression rate of those gene is relevant in colorectal carcinogenesis.Method A total of 60 colorectal samples of surgical mass resection or biopsy were collected from the Department of General Surgery of Baotou clinical medical college,Inner Mongolia medical university(Baotou Central Hospital)from March 2017 to Septembma samples and 20 er 2018.The experiment includes 20 normal colorectal samples,20 colorectal adenocolorectal cancer samples.All the 60 cases in the gene group were eligible.There were no history of radiotherapy chemotherapy and immunity therapy before surgical resection or biopsy.Immunohistochemistry was used to detect the expression of PTEN,MTDH and GATA3 in each group.The relationship between PTEN,MTDH and GATA3 in colorectal carcinogenesis was analyzed.The expression levels of these three genes were studied and correlated.Sexual analysis is applied to determine whether there is a possible regulatory relationship between them.Results(1)The positive expression rates of PTEN in three groups of different colorectal tissues were:normal tissue 100%(20/20),adenoma tissue 90%(18/20),and cancer tissue 75%(15/20).There were a statistically significant difference in the expression between normal colorectal tissues and colorectal cancer tissues(P<0.01).(2)The positive expression rates of MTDH in three groups of different colorectal tissues were: normal tissue 70%(14/20),adenoma tissue 85%(17/20),and cancer tissue 95%(19/20).There was a significant difference between the normal colorectal samples and the colorectal adenoma samples(P<0.01).There was significantly statistical difference between the normal colorectal tissue and colorectal cancer tissue(P< 0.01),the positive expression rate was different in colorectal cancer with different degree of differentiation(P<0.05).(3)There was no significant statistical difference between the age,gender,histological type and grade of intraepithelial neoplasia in PTEN+ and MTDH+ of colorectal adenoma(P>0.05).There was no significant statistical difference between the age,gender,tumor diameter,differentiation degree,depth of invasion and lymph node metastasis in PTEN+ of colorectal cancer patients(P>0.05).There was no significant statistical difference between the age,gender,tumor diameter,depth of invasion and lymph node metastasis in MTDH+ of colorectal cancer samples(P>0.05),which was correlated with the degree of differentiation of colorectal cancer patients(P<0.05).(4)GATA3 was not positively expressed in normal colorectal tissues,adenoma tissues,and cancer tissues.GATA3 did not find significant clinical significance in colorectal carcinogenesis in this experiment.Conclusions The combined detection of PTEN and MTDH is helpful to evaluate the condition of patients with colorectal cancer and has a certain interpretation effect on the early diagnosis of colorectal cancer.
