Non-alcoholic fatty liver disease(NAFLD)is a clinicopathological syndrome caused by excessive deposition of fat in hepatocytes.NAFLD is considered to be a pathological manifestation of the liver resulting from metabolic diseases such as diabetes,obesity and hyperlipidemia.Non-alcoholic steatohepatitis(NASH)is the intermediate stage of NAFLD course,in which 10%~25% patients will develop to hepatocirrhosis within10-15 years.At present,there is no effective drug for NASH treatment.Chinese traditional medicine has shown obvious advantages in the prevention and treatment of NASH.In this study,we induced the rat model of NASH by high-cholesterol-high-fat diet,and gave Sini Powder intervention to observe its influence on blood lipid,liver function and hepatic histopathological pathology in NASH rats.We hoped to expand the clinical application of Sini Powder and to provide a new means for NASH treatment.Objective: To study the hepatoprotection of Sini Powder on rats with non-alcoholic steatohepatitis and possible mechanisms.Methods: Total 32 male SD rats were randomly divided into control group,model group,high-dose and low-dose Sini Powder treatment group.Rats in control group were fed with normal food,while rats in model and treatment groups were fed with high-cholesterol-high-fat diet for 12 weeks to reproduce NASH model.From the ninth week,rats in treatment groups were given with high or low dose of Sini Powder,while rats in other two groups were given isometric normal saline.All rats were sacrificed at the end of 12 th week,serum and liver samples were collected.The total cholesterol(TC),triglycerides(TG),high-density lipoprotein(HDL),low-density lipoprotein(LDL),ALT,AST,glucose(GLU)and insulin(INS)levels in serum were measured,and the morphological changes of the liver were observed byhematoxylin-eoson(HE)staining.Results: Compared with the control group,the liver index of rats in model group increased,hepatocellular ballooning,micro-/macrovesicular steatosis occurred,accompanied by increased serum TC,LDL,GLU,ALT,AST levels and homeostasis model of insulin resistance(HOMA-IR)index.Sini Powder treatment improved the biochemical and histopathological changes in NASH rats.Conclusion: Our results suggest that Sini Powder has therapeutic potential against NASH and that it acts through multiple signaling pathways.
