Objective:To describe the expression of actin-like 6a(ACTL6a)in cholangiocarcinoma tissues and to determine the relationship of ACTL6 a with the clinicopathological characteristics and survival time of cholangiocarcinoma patients.To describe the relationship between the expression of ACTL6 a and the expression of the molecular markers of epithelial-mesenchymal transition(EMT),such as E-cadherin,N-cadherin,?-catenin,Snail and Vimentin.Determine the role of ACTL6 a plays in EMT and then help to find molecular markers for early diagnosis and drug targets for therapy of cholangiocarcinoma.In order to enhance the ability of diagnosing and treatment of cholangiocarcinoma and ameliorate the clinical outcomes of patients.Methods:The paraffin pathological specimens and case data of 50 cases of cholangiocarcinoma(experimental group)and 20 cases of normal bile duct tissue(control group)were collected.The expression of ACTL6 a was detected by immunohistochemistry.Survival data of cholangiocarcinoma patients were obtained by clinical follow-up.Analyze the relationship between ACTL6 a and the clinicopathological features of patients,such as age,gender,tissue differentiation type,TNM stage,lymph node metastasis,tumor location and the survival time.Immunohistochemistry was used to describe the different expression of EMT-related molecular markers E-cadherin,N-cadherin,?-catenin,Snail and Vimentin in cholangiocarcinoma paraffin specimens and normal bile duct paraffin specimens.And the correlation between ACTL6 a and EMT-related molecular markers were calculated.Results:1.ACTL6 a expressed in both cholangiocarcinoma and normal bile duct tissues showed by the results of immunohistochemical staining,and was mainly located on the nucleus.The positive expression rate of ACTL6 a in normal bile duct tissues was 35%,without strong positive expression,and the positive expression rate in cholangiocarcinoma tissues was 80%.Compared with normal bile duct tissue,the expression of ACTL6 a was increased in cholangiocarcinoma tissues(P<0.01).2.The expression of ACTL6 a was associated with the tissue differentiation type(P<0.05),TNM stage(P<0.05)and the metastasis of lymph node in cholangiocarcinoma patients(P<0.05).But there was no significant relationship between the expression of ACTL6 a and age,gender,or tumor location of patients.The median survival time was11.75 months in the ACTL6 a high-expression(positive)group and 30.50 months in the ACTL6 a low-expression(negative)group.Patients with high expression of ACTL6 a have a short survival time.3.Immunohistochemical staining showed that the expression of E-cadherin,N-cadherin,Vimentin,?-catenin and Snail protein was different in cholangiocarcinoma and normal bile duct tissues,and the expression of E-cadherin was decreased in cholangiocarcinoma tissues(P<0.05),while N-cadherin(P<0.05),?-Catenin(P<0.05),Snail(P<0.05),Vimentin(P<0.05)were increased.E-cadherin is mainly located on the cell membrane;N-cadherin expresses on the cell membrane and cytoplasm;?-Catenin expresses on the nucleus and cytoplasm;Snail expresses on the nucleus and cytoplasm and Vimentin is mainly located on the cytoplasm.4.In cholangiocarcinoma,the expression of ACTL6 a was positively correlated with the expression of N-cadherin(r=0.371),?-Catenin(r=0.515),Snail(r=0.509)and Vimentin(r=0.334),and was negatively correlated with the expression of E-cadherin(r=0.311),(P<0.05).Conclusion:The high expression of ACTL6 a is associated with poor prognosis and short survival time in cholangiocarcinoma patients.E-cadherin was down-regulated while N-cadherin,?-Catenin,Snail and Vimentin were up-regulated in cholangiocarcinoma and an EMT process has occurred.Moreover,the expression of ACTL6 a was negatively correlated with the expression of E-cadherin,while the expression of ACTL6 a was positively correlated with the expression of N-cadherin,?-Catenin,Snail and Vimentin.By the mechanism of promoting EMT process,ACTL6 a may lead to poor clinical outcomes of cholangiocarcinoma patients.
