OBJECTIVE Cervical cancer (Cervical cancer, CC) is one of the three most commomgynecologic tumor i,and it is a major killer of women health. At present, cervical cancertends to be younger, but the mechanism is not clear, so it is lack of methods in earlydiagnosis and treatment of cervical cancer. According to the research, wnt signaltransduction pathway is closely relation with the occurrence and development ofcervical cancer.Observing the expression of extracellular factor (wnt-1, wnt-3a) andcytoplasmic protein (β-catenin) on the gene and protein level,we explore the roles inthe development of cervical cancer,which can infer a new basic theoretical for theoccurrence and development of cervical cancer,provide a new biological index todiagnosis SCC earlier and find a new idea for target treatment of cervical cancer.METHODS (1) The expression of wnt-3a and β-catenin were detected bySP-immunohistochemical method in20cases of normal cervical tissues,24cases ofcervical intraepithelial neoplasia tissues and56cases of squamous carcinoma of thecervix tissues.The testing results were analyzed by statistical analysis according to thedata.(2) Total RNA was extracted from normal cervical tissues, cervical intraepithelialneoplasia and squamous carcinoma of the cervix tissues.The mRNA expression ofwnt-1and wnt-3a was detected in different tissues by RT-PCR method.RESULTS (1) the expression of wnt-3a protein in different cervix tissues Thepositive expression rate of wnt-3a in normal cervical tissues, CIN tissues and cervicalcarcinoma tissues were10%(2/20),37.5%(9/24) and39.3%(22/56) respectively.and there was significant difference between3groups (χ2=6.006, P<0.05).In different clinical stages of cervical cancer, the expression of wnt-3a in I phase, IIphase and III phase had no statistical difference (P>0.05).The positive expression rateof wnt-3a was increased with the deepens of pathological grading degree and infiltration,but the difference was not statistically significant (P>0.05).(2) the expression ofβ-catenin protein in different cervix tissues The expression of β-catenin protein isshowing continuous membrane expression in normal cervical squamous epithelialtissues.In CIN and SCC tissues,the expression of membrane is showing differentdegrees of loss or no expression. The β-catenin protein express abnormally in thecytoplasm and cell nucleus. The abnormal expression rate of β-catenin protein in normalcervical tissues, CIN tissues and SCC tissues were15%(3/17)、50%(12/24)and82.1%(46/56)respectively, and there was significant difference among3groups (χ2=29.532,P<0.01).With the pathological grade of cervical cancer increasing,the abnormalexpression of β-catenin gradually increased, and there was significant difference among3groups (χ2=8.883, P<0.05)..In the shallow invasion,the abnormal expression rate ofβ-catenin protein was66.7%(20/30), and in deep infiltration group, the abnormalexpression rate of β-catenin protein was100%(12/12).There was significant differencebetween2groups(P>0.05).but it is not related to clinical stages.(3) in56cases ofcervical cancer tissues,the positive expression of wnt-3a and the abnormal expression ofβ-catenin at the same time was totle20cases,and the negative expression was8cases,there was positively correlated with the expression of β-catenin and wnt-3a(r=0.181).(4),from the normal cervical tissues to SCC, the expression of wnt-1andwnt-3a mRNA increased gradually.CONCLUSIONS1.The wnt signal pathway activation and the abnormal expression ofβ-catenin were closely related to the occurrence and development of cervicalcancer,and it may be used as an additional criterion for early diagnosis of. cervical cancer.2.Beta-catenin protein was closely related to the depth of invasion.and malignantdegree of tumor.3.The combined detection of wnt-3a protein and-catenin protein may be diagnosisearlier and judge the malignant degree of ervical cancer more accurately.
