Objectives To investigate the mechanism of suppressor gene SASH1 inhibiting breast cancer metastasis and the role of Notch1 signaling pathway in it.Methods The established human SASH1 eukaryotic expression vector pcDNA 3.1(+)-SASH1 was transferred into human breast cancer cells and the SASH1 stable expression cell line of human breast cancer with excessive expression of the SASH1 was established by G418 screening.The transfection empty carrier was used as a control group.RTqPCR and Western Blot were used to verify the expression levels of mRNA and protein of SASH1 in stable expression cell lines.Western blot and RT-qPCR were used to detect the expression of Notch1 related genes in cell signaling pathways to determine their activation status.The ability of cell migration in expression group and control group was tested by cell scratch experiment.The effects of SASH1 expression on the survival of breast cancer patients were studied using the Kaplan-Meier database.Results The stable expression cell lines of breast cancer MCF 7 and MDA-MB231 expressing human SASH1 were successfully obtained.There was no significant difference in cell migration ability of breast cancer in overexpression group and control group.In 48 hours,the distance of the high expression group of SASH1 was wider than that of the control group(`x =1.4mm and`x =1.7mm respectively p<0.05).After SASH1 was expressed,the expression of Notch1 decreasedunder SASH1 overexpression indicatliy that SASH1 could inhibite the pathway.The Kaplan-Meier database showed that the lifetime of the SASH1 expression group was lower than that of the expression group(HR = 0.78).Conclusions The SASH1 could inhibit breast cancer metastasis by regulating the Notch1 signaling pathway.This study here would provides new experimental data for elucidating the role of SASH1 in breast cancer mechanism.Figure 4;Table 0;Reference 74.
