Objectives Study on the expression and Regulation of miRNA-1290 and ARL4 C in placenta of patients with preeclampsia.Methods 1 63 cases of placental tissue(29 cases of normal group,14 cases of preeclampsia group,preeclampsia severe group 20 cases)were collected from cesarean section delivery in Tangshan Workers' Hospital from November 2017 to June 2018.Realtime fluorescence quantitative polymerase chain reaction(RT-PCR)was used to detect the expression of miR-1290 in placenta of the three groups,and Western blot was used to detect ARL4 C protein expression differences.2 JEG-3 cells were cultured and transfected into blank group,transfection reagent group,miR-NC group and miR-1290 mimics experimental group by cell transfection technology.The expression levels of ARL4 CmRNA in each group were detected by RT-PCR technology,and the expression of ARL4 C protein in each group was detected by Western blot method.CCK-8 technology was used to detect the proliferation ability of cells,and scratch test was used to detect the invasion ability of cells.3 Using SPSS 17.0 software,the measurement data were expressed as mean ±standard deviation(x ±s).T test was used for the comparison of the two samples,and single factor analysis of variance or rank sum test was used for the comparison of multiple samples.There was significant difference is P < 0.05.Results 1 The relative expression levels of miR-1290 in placental tissues of normal group,preeclampsia group and severe preeclampsia group were 1.015±0.030,1.256±0.425 and 1.797 ± 0.049,respectively,showing an upward trend,with statistically significant differences(F=2307.861,P< 0.05);The relative expression levels of ARL4 C protein in placental tissues of the three groups were 0.769 ± 0.375,0.528 ±0.462,and0.290±0.312,respectively,showing a decreasing trend(F=959.758,P < 0.05).2.After the successful transfection of JEG-3 cells,the expression levels of ARL4 CmRNA and ARL4 C protein in the miR-1290 mimicis transfection group were lower than other three groups(F=123.008,F=37.303,P < 0.05).3.CCK-8 results showed that the OD values of the blank group,the transfection reagent group,the miR-NC group and the miR-1290 mimics group were 0.51 ± 0.02,0.53 ± 0.00,0.53 ± 0.00 and 0.41 ± 0.01,respectively.The miR-1290 mimics group showed statistically significant differences compared with the other three groups(F=94.743,P < 0.05).The relative values of scratch area change rate(48h)in the four groups were 61.03 ± 1.53,57.13 ± 5.12,54.10 ± 4.31 and 31.00 ± 6.63.The difference between the experimental group and the other three groups was statistically significant(F= 24.041,P < 0.05).Conclusions 1 ARL4 C is the target gene of miR-1290.2 In the occurrence and development of preeclampsia,miR-1290 with high expression is involved in the process of disease by negatively regulating the expression of ARL4 C,both of which play an important role.3 miR-1290 can lead to the occurrence of preeclampsia by affecting the proliferation,infiltration and migration of trophoblast cells.4 miR-1290 may be a target for the prevention or treatment of preeclampsia.Figure12;Table16;Reference 159.
