| Objectives Through the establishment of an animal model of osteoporosis rat dry trough,drug intervention of alendronate was performed during the experiment to observe the healing of alveolar fossa in osteoporotic rats after dry trough and to detect osteoprotegerin(OPG),expression of tartrate-resistant acid phosphatase(TRAP).To explore the effect of alendronate on the healing of alveolar fossa in rats with osteoporosis and osteophyte,and provide experimental evidence for the treatment of dry trough in patients with clinical osteoporosis.Methods Fifty-four healthy 12-week-old SD rats weighing 200-250 g,SPF grade,were fed with sterile mouse pellets.The animals were given free access to drinking water.After one week of adaptive feeding,the rats were observed to be stable and normal.All rats were randomly divided into three groups,namely: control group,osteoporosis dry socket model group,osteoporosis dry socket + alendronate treatment group;18 in each group.Except the control group,the other groups were treated with 100 mg/(kg·d)of retinoic acid by intragastric administration;the control group was given the same dose of distilled water for gavage treatment for 14 days.After 14 days,the femur bone density of each group of rats was examined to determine the establishment of an osteoporosis model.Then,the maxillary left first molars of each group of rats were removed,and the dry trough model was established by using adrenaline to stop bleeding and contaminating the tooth extraction.Except for the daily intervention of sodium alendronate(1.05 mg/kg)for drug intervention in the treatment group,the other two groups were given the same daily dose of distilled water.Rats were anesthetized at 1,2,and 4 weeks after successful establishment of the osteoporosis dry trough model.The bone tissue of the maxillary extraction socket was taken out for HE staining,osteoprotegerin(OPG)immunohistochemistry and tartrate-resistant acid phosphatase(TRAP)-specific staining.The results were expressed as mean ± standard deviation(sx±),and one-way analysis of variance was used for comparison between groups.When P<0.05,the difference was statistically significant.Results 1 After two weeks of retinoic acid treatment,the bone mineral density of the femur in the model group and the alendronate sodium treatment group was significantly lower than that in the control group(the difference was statistically significant(P<0.05).2 After 3 days of tooth extraction,the wounds of the rats were seriously polluted,and the obvious purulent secretions were accompanied by rancid taste.3 Histological observation: The alendronate sodium drug treatment group had faster inflammation regression than the control group and the model group,and the new trabecular bone and new bone in the treatment group in the same period had good healing effect.4 OPG test results: During the 7d,14 d,and 28 d periods,the positive expression of the alendronate sodium treatment group was higher than that of the control group and the model group,and the difference was statistically significant(P<0.05).5 TRAP test results: During the 7d,14 d,and 28 d periods,the positive expression of the alendronate sodium treatment group was lower than that of the control group and the model group(P<0.05).Conclusions 1 The model of osteoporosis can be successfully established by continuous intragastric administration of retinoic acid for 2 weeks,and the dry trough model can be established by applying adrenaline and contaminating the extraction.2 Experiments have shown that rats with osteoporosis and dry trough have poor healing,and osteoporosis can lead to delayed healing of dry trough.3 Alendronate sodium can increase the expression of OPG in osteoporosis dry trough rats,reduce the expression of TRAP,and promote the healing of extraction of osteoporosis in rats with osteoporosis.Figure 36,Table 5,Reference 149... |
