ObjectivesThe mice were exposed to bisphenol A?BPA?and the effects of bisphenol A on the ovary and uterus of mice were investigated.Methods Thirty Kunming 6?KM?female mice were randomly divided into control group?corn oil?,low-dose group?BPA100mg/kg?,high-dose group?BPA200 mg/kg?,a total of 3groups,each Group 10 only.The bisphenol A solution was administered by intragastric administration.The dose was 0.01 ml/g,once a day,5 days per week,and continuous exposure for 12 weeks.1 General conditions and weight:observe the mental state of the mouse,fur gloss,activity,diet,drinking water and body weight changes;2 after the last exposure for 24 hours,the eyeballs were taken and the mice were sacrificed by cervical dislocation.The mouse ovary and uterus tissue were quickly taken out and weighed separately to calculate the organ coefficient.3 The pathological changes of mouse ovary and uterus were observed under microscope.4 After the eyeball was taken,the blood was centrifuged and the serum was collected.Enzyme-linked immunosorbent assay?ELISA?was used to determine the levels of estradiol?E2?,progesterone?P?and follicle stimulating hormone?FSH?in mouse serum;5 to prepare mouse cell ovary tissue for single cell suspension using fluorescence detection Oxygen cell reactive oxygen species?ROS?levels were determined by DCFH-DA method and flow cytometry;6 mouse ovarian tissue was dissected to prepare ovarian tissue suspension,and total antioxidant capacity in mouse ovarian tissue was determined by Fe3+reduction method?T-AOC?,spectrophotometric determination of glutathione?GSH?content in mouse ovarian tissue,determination of malondialdehyde?MDA?content in mouse ovarian tissue by thiobarbituric acid method;7preparation of mouse ovarian tissue disease The section of the ovarian cells was determined by TUNEL method,and the apoptosis rate was calculated.Results 1 Compared with the control group,the mice in the BPA-treated group had a weaker mental state,dull skin,slower movement,and reduced diet and drinking water.At the 6th and 12th week of exposure,the weight of the low-dose group?36.11±0.34?g,?37.40±0.80?g and high-dose group weight?33.49±1.28?g,?34.87±1.04?g were lower than exposure can affect the general condition and body weight of mice,among which high dose of BPA is more obvious;2 BPA exposure can cause wet weight of ovary in mice,and the number of follicles at all levels is reduced;low dose of BPA can cause wetness of mouse uterus The weight of the uterine cavity is reduced,and the high dose of BPA will increase the wet weight of the uterus and the area of the uterine cavity;3BPA exposure will lead to a decrease in the serum E2,P and FSH levels,and the high dose of BPA.More obvious;4 BPA exposure can lead to decreased T-AOC activity,GSH content,increased ROS and MDA content in mouse ovarian tissue,and higher dose of BPA is more obvious;5 BPA exposure leads to mouse ovarian cells The apoptotic rate is increased,and the effect of high dose of BPA is
