Objectives To study the therapeutic effect of reduced glutathione(GSH)on severe acute pancreatitis and the expression of endoplasmic reticulum stress proteins GRP 78 and Caspase-12.Methods Firstly,Sixty adult male Sprague-Dawley rats,10 days after standard feeding,were randomly divided into the simple switch abdominal group(Sham group),the severe acute pancreatitis group(SAP group)and the reduced glutathione treatment group(GSH group).3 groups(N=24),in which Sham group was anesthetized and opened the abdominal cavity to turn the pancreas,then the abdomen was closed.GSH group was intraperitoneally injected with GSH(1g/kg)2 hours before model establishment,then the pancreaticobiliary tube was exposed open,and the bovine sulphate was slowly injected.Sodium cholate(5g/kg),the abdomen was closed after successful modeling,and the same dose of normal saline was intraperitoneally injected 2 hours before the SAP group was established.The modeling process was the same as that of the GSH group.Rats were reopened at 3 h,6 h,12 h,and 24 h after successful manufacturing.The pancreas was removed and HE stained.The histological changes of pancreas in each group were observed.The inferior vena cava was exposed to open laparotomy.The changes of serum amylase and IL-6 and C-reactive protein in rats were obtained.The expression of GRP 78 and Caspase-12 in pancreas of each group were detected by immunoblotting.Results 1.Histopathological findings of pancreas showed that after laparotomy,no obvious hemorrhage and edema were observed under the naked eyes of the pancreas.The pancreatic tissue was normal under light microscope,no hemorrhage and inflammatory cells infiltrated and aggregated;pancreatic hemorrhage and necrosis were observed in the SAP group.Under light microscopy,pancreatic cell necrosis and hemorrhage can be seen,scattered red blood cells can be seen in the tissue,and inflammatory cells mainly infiltrated by neutrophils can be seen,and the longer the time after modeling,the more severe the inflammation;the pancreatic tissue under the light microscope of GSH group The degree of inflammation was reduced compared with the SAP group,and the inflammation was aggravated with prolonged modeling time.2.Serum amylase,IL-6 and C-reactive protein results showed that serum amylase,IL-6 and C-reactive protein were detected at the same time point,and serum amylase,IL-6 and C-reactive protein levels were observed at the same time point in the SAP group.Compared with the GSH group,the difference was statistically significant(P<0.05).There was no significant change in the levels of amylase and IL-6 and C-reactive protein in the sham group(P>0.05).3.SAP group and GSH group The expression levels of GRP 78 and Caspase-12 increased,and the expression level of SAP histones was higher than that of GSH group at the same time point.Conclusions Reduced glutathione can exert its therapeutic effect on severe acute pancreatitis in rats by reducing the expression levels of GRP 78 and Caspase-12.Figure 3;Table 5;Reference 161.
