Objective To study the expression of cyclooxygenase-2(COX-2) and nuclear factor-kappa B(NF-κB) in rats with L-arginine(L-Arg)-induced acute pancreatitis (AP). And to assess the effctiveness of Reduced Glutathione Sodjum(GSH) on the activity of them and on the pathological severity.Methods Ninety-six Spraque-Dawley male rats were divided into three groups randomly. AP were induced by injected intraperitoneally three times with 6% L-Arg (3×1.0mg/g) in an interval of 1 hour in group A and group G. Group C received saline alone. Group G was treated with 10% GSH (0.6mg/g) at 1 hour before the L-Arg induced. Rats were killed at 2nd,6th,12th and 24th hour after the last time of L-Arg injection. The expressions of COX-2 and NF-κB in the pancreatic tissue were tested with the method of immunohistochemistry. The serum levels of amylase were assayed pathologic changes of pancreatic tissue were scored according to Rongione's standard.Results Compared to group C,serum levels of amylase in group A and G were significantly increased at different time(P<0.01).But compared to group A, serum levels of amylase were significantly decreased in group G (P<0.01).Pathologic changes of pancreatic tissue in group G were significantly decreased than group A at different time(P<0.05 or P<0.01).There were few of COX-2 and NF-κB expressed in group C. The two cytokines were activated in group A and G in early period, and reach its peak at 12th hour, than decrease.
