Objective To explore the effect and mechanism of up-regulation of the biological rhythm gene Cry2 on the function of MG63 in osteosarcoma cells.Method1.Lentioma was transfected into osteosarcoma MG63 cells,and Cry2 up-regulated(MG63-Cry2U)and negative control(MG63-con)stably transfected cell lines were constructed.Cry2 m RNA and protein expression levels were examined to confirm the successful establishment of Cry2 up-regulated stable transfected cells.2.Cell functions test: The changes of osteosarcoma MG63 cell functions were testeded after up-regulation of Cry2.3.Explore signaling pathway-related genes expression level after Cry2 overexpression.Result1.Cry2 overexpressed stably transfected cells in osteosarcoma MG63 was successfully established.2.Compared with MG63-con,osteosarcoma cell MG63-Cry2 U has weakened proliferation,invasion and migration ability,increased proportion of apoptotic necrosis,and increased proportion of cell cycle G1 phase.3.After up-regulating Cry2,the expression of P53 was increased,and the expression of C-myc,Cyclin D1 and MMP-9 was decreased.ConclusionOverexpression of Cry2 expression can inhibit the proliferation and invasion and migration of osteosarcoma cell line MG63,promote apoptosis and necrosis,and induce cell cycle G1 arrest.Cry2 may be a tumor suppressor gene in osteosarcoma. |
