Objectives:Development and Application of a Biological Activity Evaluation Method Based on Anti-influenza Virus Neuraminidase Reagent Kit for the Clinically Commonly Used Drugs of Radical Detoxification: Radix Isatidis,Coral Coral,and Lianhua Qingling CapsuleMethods:1 Inspected the stability,reproducibility and consistency of the influenza virus neuraminidase inhibitory titer assay kit;2 Standardization and application of quality assessment techniques for anti-NA activity of Radix Isatidis;3 Quality assessment of anti-NA activity of Sarcoderin Application;4 Lianhua Qingqi capsules anti-NA activity quality evaluation technology standardization and application.Results:1.The stability,reproducibility and consistency of the test kits for influenza virus neuraminidase inhibition titers were examined;the average inhibition rate of intra-day precision for the same sample was 85.34%,RSD <1.3%,and the average inhibition rate of daytime precision.85.39%,RSD <2.5%,repetitive average inhibition rate was 85.42%,RSD <3.15%.The average blank OD value of the six freeze-thaw cycles of the same kit was 11239.96,and the RSD was less than 5.8%.The average OD of the same kit was six freeze-thaw cycles and the RSD was less than 12.75%.The average inhibition rate of six freeze-thaw cycles of the same kit was 86.61%.,RSD<1.70%.It is recommended that the kit be used within six freeze-thaw cycles.Six laboratory reproducibility experiments were conducted and there was no significant difference between the trials.Through research,it has been found that this method has good precision and repeatability and can be used to evaluate the inhibition of influenza virus activity by drugs.On this basis,a standard operating procedure for the determination of influenza neuraminidase inhibition titer kit was established.2 Standardization and application of quality assessment of antibacterial activity of Radix Isatidis: Banlangen is a commonly used heat-clearing and detoxifying traditional Chinese medicine,which has the activity of inhibiting influenza virus.10 batches of Radix Isatidis medicinal materials and 20 batches of Banlangen granules from different manufacturers were determined and evaluated chemically.The determination of biological valence and the establishment of a standardized system for the quality evaluation of Radix Isatidis are the weakest links in its research.This is based on the glucosinolate-glucosidase system as the main target.The chemical composition and anti-influenza virus effect of the blister and its enzymatic hydrolysates laid a chemical foundation for the elucidation of the potent material basis of Radix Isatidis and opened up new directions for development.3 Quality assessment of anti-NA activity of grass corals Technical standardization and application: Caoshanhuo has the functions of antibacterial,anti-inflammatory,heat-clearing,detoxifying,and bone-building.It has antibacterial and anti-inflammatory effects,inhibits influenza virus,promotes fracture healing and anti-tumor biological activity.Ten batches of grass corals were established for chemical composition determination and characteristic maps.Based on the results,influenza virus neuraminidase inhibition titer detection kit was used for detection,and the biological titer of each batch was determined.A standardized system for quality assessment of grass corals.4 Standardization and application of quality evaluation of anti-NA activity of Lianhua Qingqi Capsules: 10 batches of Lianhua Qingqi capsules were collected for the determination of chemical composition and the establishment of characteristic maps.The biological titer of each batch was determined.Established a standardized system for quality assessment of Lianhua Qingqi Capsules.Conclusion:Based on the previous studies,this topic has optimized the biological evaluation kit based on the detection of influenza virus neuraminidase antiviral activity,and established a standardized system for the quality assessment of Radix Isatidis,Coral Coral,and Lianhua Qingchao Capsules.It lays a foundation for clearing up heat and detoxifying the traditional Chinese medicine to establish a standardized system of quality assessment technology centered on biological evaluation.
