Objective To observe the effect of Compound Opening Arrow Mixture on the growth and apoptosis of mouse breast cancer 4T1-luc and the expression of apoptosis protein Caspase-3 and Cle-caspase-3.Methods Luciferase-labeled mouse breast cancer cells 4T1-Luc were inoculated into the mouse breast pad to establish a luciferase-labeled mouse breast cancer cell model.Sixty BALB/C-nu mice were randomly divided into normal controls group,model group,Compound Opening Arrow Mixture(high,medium,and low dose)groups,low-molecular-weight heparin group,and 10 each.Except that the normal controls group was not modeled,all the mice in the other groups were given a drug intervention after successful modeling.The group of Compound Opening Arrow Mixture(high,medium and low dose)was given 6g/ml,3g/ml,1.5g/ml Compound Opening Arrow Mixture Chinese medicine;low-molecular-weight heparin group was given low molecular weight heparin(1500U/ Kg)intraperitoneal injection;the model group and the normal controls group were given an equal dose of distilled water.The number and distribution of transplanted tumors in 4T1-luc breast cancer cells were observed in nude mice by in vivo imaging system,at the time of inoculation,after successful modeling,and on the 7th and 14 th day after administration.Tumor cell apoptosis was detected by TUNEL method;Caspase-3 m RNA expression was detected by RT-PCR;Caspase-3 and Cle-caspase-3 protein expression was detected by Western Blot.Results The in vivo imaging system showed that the total photon number in each treatment group was significantly lower,and Compound Opening Arrow Mixture low-dose group> medium-dose group> high-dose group.The apoptosis index of Compound Opening Arrow Mixture was detected by TUNEL,and the apoptotic index increased with the increase of dose.And the 14 th day> the 7th day.The expression of Caspase-3 m RNA in each treatment group was significantly increased by RT-PCR,and the expression level of Compound Opening Arrow Mixture high-dose group> medium-dose group> low-dose group.The expressions of Caspase-3 and Cle-caspase-3 in the high,medium and low groups of Compound Opening Arrow Mixture were significantly increased by Western Blot,showing a dose-and time-dependent manner.By comparison,it was also found that the total photon number of the high-dose group of Compound Opening Arrow Mixture was lower than that of the low-molecular-weight heparin group on the 14 th day after administration,and the expression of apoptotic index,Caspase-3 m RNA,Caspase-3 and Cle-caspase-3 protein were higher than the low-molecular-weight heparin group.Conclusion “Compound Opening Arrow Mixture” can inhibit the growth of 4T1-luc breast cancer xenografts in mice,and exhibit dose tolerance and time tolerance.“Compound Opening Arrow Mixture” has the effect of up-regulating the expression of Caspase-3 protein and promoting apoptosis of breast cancer cells.
