OBJECTIVE: Primary cilia participate in various signal transduction,suggesting a potential role in cancer development.OFD1 located in the centriolar satellites,is able to inhibit the ciliogenesis.Induced of autophagy degraded OFD1 in the satellites and dramatically increased cilia formation.The role of primary cilia playing in uveal melanoma is unclear.Here we aim to induce ciliogenesis in UM and explore the function and mechanism of primary cilia in tumor progress.CONTENT AND METHODS: 1.Primary cilia,basal autophagy and OFD1 were estimated by IF,real time PCR and Western blot in UM cells.2.Induction of autophagy increased cilio formatio n in UM cells,and Stable cell lines,CCK8 assay,cell cycle and transwell assay were performed to estimate the influence of OFD1 on tumor characters.3.Monitor the change of the expression of OFD1 when autophagy is activated.CCK8 assay,cell cycle,and transwell assay were performed to estimate the influence of primary cili a on tumor characters.CONCLUSION: 1.Few primary cilia,low basal autophagy and high expression of OFD1 in UM cells.2.Autophagy in UM cells was activated which formed primary cili a and degraded OFD1 protein.Primary cili a inhibited proliferation,migration of UM cells.3.Depletion of OFD1 inhibited tumor growth but didn't change migrationACHIEVEMENT: Activated autophagy degraded OFD1 in satellites and formed primary cilia in UM,which inhibited tumor growth and migration.OBJECTIVE: Long non-coding RNAs(lnc RNAs)have been appreciated as an integral component of gene regulatory networks,by regulating the protein-coding genes,and also suggest a potential link to many human diseases.Retinoblastoma is the most common primary tumor in eyes,and always affects children.It can lead to incurable invasion of the optic nerve and brain.Here we aim to find lnc RNA in RB and explore the function and mechanism of lnc RNA in tumor progress.METHODS 1.The expression of CUM1 in RB cells and the construction of stable cell lines.2.Assays were performed to estimate the influence of lnc RNA on tumor characters.CONCLUSION CUM1 is a novel lnc RNA which functions as a tumor suppressor and inhibits proliferation,of tumor cells.ACHIEVEMENT We defined a novel lnc RNA which serves as a tumor suppressor.
