Objective Screening the key lncRNA in UM cell and how lncRNAs operate in the regulation of autophagy in the Uveal melanoma.Contect and methods The mTOR inhibitors such as rapamycin?PP242?Torin1 can induce autophagy in cells.The results were proved by western blot,Cell immunofluorescence and GFP-LC3 fusion protein.In UM,Cell autophagy decreased the ability of proliferation in cells by MTT assay.After being imposed by PP242 48 h or 72 h,cell autophagy cause G1 phase retardation.Heap map displays the lncRNAs differentially expression.Overexpression of lncRNA-TC01005809 can increase the autophagy level of UM cells.And,Overexpression of lncRNA-TC01005809 decreased the ability of metastasis Conclusion The mTOR inhibitors can induce autophagy in cells.lncRNA-TC01005809 can increase the autophagy level of UM cells and decrease the ability of metastasis.Achievement In UM cells,lncRNA-TC01005809 can increase the autophagy level and decrease the ability of metastasis.It provides clues for the future study.
