Skin trauma is a common clinical problem in plastic surgery.Diabetic skin injury in chronic wounds is an important cause of refractory wound.The treatment of scalp avulsion injury in acute skin trauma is particularly critical,and it is a great challenge to clinicians when it is combined with vertebral injury.This study is to explore the role of endoplasmic reticulum stress in the skin lesion of diabetic rats and characteristics of scalp avulsion injury combined with vertebral injury.Objective:It aims to evaluate whether the different concentration of glucose can cause the occurrence of endoplasmic reticulum stress in rat skin fibroblasts culture.When the model of skin lesion in diabetic rats is established,we need to observe the basic pathological changes and the level of endoplasmic reticulum stress in the diabetic rat skin lesion.It is aimed to the incidence and characteristics of scalp avulsion combined with vertebral body injury.Materials and methods:With diabetic skin lesion model,we detect the basic pathological changes,gene expression of endoplasmic reticulum stress.In vitro,It is to observe changes in gene and protein expression of endoplasmic reticulum stress in rat skin fibroblast culture with different glucose concentrations.Patient history of scalp avulsion injury in the past 20 years is be explored in the medical record office.Results : In vitro,it is showed that higher glucose concentrations can lead to higher endoplasmic reticulum stress in rat skin fibroblast culture.In vivo,the skin lesion is associated with the endoplasmic reticulum stress,which leads to induce apoptosis.Among the skin lesions(the scalp avulsion injury)64 patients,9 patients are found to be involved in vertebral injury.Conclusions : Higher glucose may cause the skin fibroblast injury with higher the endoplasmic reticulum stress level.In the skin lesion of diabetic rats,the expression of apoptosis gene of the endoplasmic reticulum stress in diabetic rats is much higher than the expression in the control.It may be a new target for clinical treatment to diabetic skin lesion.Attention should be paid to the existence of vertebral injury in acute injury of scalp avulsion.
