Objective: To study the effects of ethyl acetate extract of hematoxylin on serum levels of tumor necrosis factor-?(TNF-?),monocyte chemokine-1(MCP-1)and pathological morphology of myocardium in diabetic rats.Methods: Choose 24 healthy male SD rats,adaptive feed after 1 week,randomly selected from 6 as blank control group,will the remaining 18 rats fasting,forbidden by intraperitoneal injection of water after 12 hours to be single STZ 50mg/kg,diabetic rats model was established,the 18 diabetic model rats were randomly divided into model control group,?-lipoic acid,and hematoxylin group,each group of six.Blank control group and model control group were given carboxymethyl cellulose sodium suspension by gavage,?-lipoic acid group by intraperitoneal injection of 20 mg/kg/d of?-lipoic acid,and sapphire group by gavage of ethyl acetate extract suspension of hematoxylin,equivalent to 1.15 g/kg/d of raw drug,once a day for 8 weeks.Blood samples were collected after 8 weeks.Changes in TNF-?and MCP-1 levels in serum of rats in each group were detected and compared by elisa.The structure of rat cardiac tissue was observed by HE staining.Results: 1.TNF-?: Compared with blank control group,model control group rats serum TNF-? levels increased significantly,the difference is statistically significant(P<0.01),compared with blank control group,?-lipoic acid,hematoxylin group rats serum TNF-? content is slightly higher,there was no statistically significant difference((P>0.05),compared with model control group,?-lipoic acid,hematoxylin group rats serum TNF-?content is significantly reduced,statistically significant difference(P<0.01),?-lipoic acid,and hematoxylin group rats serum TNF-?content in comparison,there was no statistically significant difference(P>0.05).2.MCP-1: compared with blank control group,model control group increased MCP-1 content in serum,statistically significant difference(P<0.01),compared with blank control group,?-lipoic acid,hematoxylin group rats serum MCP-1 content is slightly higher,there was no statisticallysignificant difference((P>0.05),compared with model control group,?-lipoic acid,hematoxylin group MCP-1 content in serum of rats decreased,statistically significant difference(P<0.05),?-lipoic acid,and hematoxylin group MCP-1 content in serum of rats,there was no statistically significant difference(P>0.05).3.Results of HE staining of myocardial tissue:(1)Blank control group:normal myocardial tissue,orderly arrangement of myocardial cells,clear structure,normal cell morphology.(2)Model control group: myocardial tissue was severely damaged,myocardial cells were disordered and disordered,and a large number of cells were swollen and congested,accompanied by a large number of inflammatory cells infiltration.(3)?-lipoic acid group: myocardial tissue was slightly damaged,myocardial cells were slightly disordered,with slightly irregular arrangement,a small amount of cell swelling and congestion,and a small amount of inflammatory cell infiltration.(4)Hematoxylin group: myocardial tissue was slightly damaged,myocardial cells were slightly disordered,with slightly irregular arrangement,a small amount of cell swelling and congestion,and a small amount of inflammatory cell infiltration.Conclusion: 1.Ethyl acetate extract of hematoxylin can reduce the levels of TNF-? and MCP-1 in serum of diabetic rats,and inhibit the inflammatory state in diabetic macroangiopathy.2.Ethyl acetate extract of hematoxylin can improve the pathomorphological changes of myocardial tissue in diabetic rat models and play a protective role in the myocardial tissue of rats.
