| Objective To study the effect of Fu Zheng Hua Yu Jiang Zhuo Tong Luo Recipe on renal function,renal patho-logy and the expression of VEGF and tsp-1 in renal tissue of5/6 nephrectomy rats,and to further explore its mechanism of delaying renal fibrosis.Methods109 male SD rats,weighing 180-220 g,were selected for adaptive feeding for one week.Then they were randomly divided into sham operation group,model group and traditional Chinese medicine group(low,medium and high dose groups).Except sham-operated group,the other four groups underwent 5/6 nephrectomy.Then the intervention began after one week of postoperative feeding.The model group was given saline and the traditional Chinese medicine group was given low,medium and high doses of traditional Chinese medicine respectively,during which the general situation,diet and drinking water quantity,mental behavior state and weight change of the rats were observed.Rats were sacrificed at 4,8 and 12 weeks after intragastric administration?Blood samples were taken from the abdominal aorta and used to determined the serum creatinine and urea nitrogen after isolated serumand.Residual kidney tissue was collected and paraffin sections were used for HE,Masson and PASM staining to observe the degree of renal lesions.The expression of VEGF and tsp-1 in kidney tissue of 5/6 nephrectomy rats was analyzed by immunohistochemical semi-quantitative analysis.The experimental data were expressed by` x±s.And SPSS23.0 was used for one-way ANOVA analysis of variance between groups,and comparisons between groups were tested by independent sample T test,and used to Pearson test analyze the correlations,if P < 0.05,indicated that there was statistical significance.Results?.The Effect of Fuzheng Huayu Jiangzhuotongluo Recipe on General Conditions of 5/6 Nephrectomized Rats The rats in the sham operation group were generally in good condition,They lived a normal life.In the model group,the mental state of the rats gradually deteriorated,with rough,dark and yellow hair,loss of luster,decreased mobility,irritable,and reduced food intake.Some rats gradually developed such phenomena as foam urine,hematuria,oliguria,and loose stool.The general condition of the rats in the TCM group was worse than that in the sham group,but the above condition was obviously better than that in the model group.?.The Effect of Fuzheng Huayu Jiangzhuo Tongluo Recipe on Serum Creatinine and Urea Nitrogen in Rats in 5/6 Nephrectomy Serum creatinine and urea nitrogen in sham-operated group maintained a relatively stable level.Compared with the sham operation group,serum creatinine and urea nitrogen levels in the model group increased slowly,with significant difference(P < 0.01).Compared with the model group,the serum creatinine level in the traditional Chinese medicine group was significantly lower than that in the model group,and the difference was statistically significant(P < 0.05),and the effect was the best in the high-dose traditional Chinese medicine group.?.The Effect of Fuzheng Huayu Jiangzhuotongluo Recipe on the Degree of Renal Fibrosis in 5/6 Nephrectomized Rats There were no obvious pathological changes in the kidneys of rats in sham-operated group.Compared with the sham group,the area of renal fibrosis in the model group was gradually enlarged with the extension of time under HE staining.Compared with the model group,the area of renal fibrosis was decreased in each group of traditional Chinese medicine.The scores of interstitial fibrosis of renal tubules under MASSON staining and glomerulosclerosis index under PASM staining were significantly higher in the model group than in the sham group(P < 0.01),and significantly lower in the traditional Chinese medicine group than in the model group(P < 0.01).There was no statistical significance between the groups of Chinese medicin?.The Effect of Fuzheng Huayu Jiangzhuotongluo Recipe on the Expression of VEGF and TSP-1 in Renal Tissue of 5/6 Nephrectomized Rats In the sham-operated group,the expression of vascular endothelial growth factor was widespread in glomerular podocytes and tubular epithelial cells.The expression of vascular endothelial growth factor in the glomeruli and tubules of the model group decreased gradually with the prolongation of time.Compared with the sham operation group,the expression of vascular endothelial growth factor had statistical significance(P < 0.01).The expression of vascular endothelial growth factor in the 4-week Chinese medicine group was widespread,but there was no statistical significance compared with model group(P > 0.05).The expression of vascular endothelial growth factor in the 8-week and 12-week Chinese medicine group was higher than that in the model group(P < 0.05),which was statistically significant.The expression of TSP-1 in the glomeruli and tubules was minimal in the sham-operated group.The expression of TSP-1 was observed in the model group,and increased gradually with time,which was significantly different from that in the sham operation group(P < 0.01).Compared with the model group,the expression of TSP-1in the traditional Chinese medicine group was decreased(P < 0.05).?.The Analysis Correlation between the Expression of VEGF and TSP-1 with Renal Fibrosis The correlation analysis showed that the expression of vascular endothelial growth factor was negatively correlated with serum creatinine,renal fibrosis and TSP-1,while the expression of TSP-1 was positively correlated with serum creatinine and renal fibrosis.Conclusion Fuzheng Huayu Jiangzhuo Tongluo Recipe can improve the general condition of 5/6 nephrectomized rats,reduce the degree of renal fibrosis and delay the progress of renal fibrosis.Its
