| Objective:Visual impairment caused by diabetic retinopathy(DR)has become an important factor affecting the quality of life of diabetic patient.At present,many studies have found that the pathogenesis of DR is related to RhoA/ROCK pathway and intercellular adhesion molecule-1(ICAM-1)and vascular cell adhesion molecule-1(VCAM-1).Most of the above studies are cell and animal models,and the reports on serum RhoA icam-1 vcam-1 expression levels in healthy people with type 2 diabetes and DR patients are rare.In this study,the expression levels of RhoA,ICAM-1 and VCAM-1 in serum of the healthy control group,the simple diabetes group and the DR group were observed,and the relationship between the changes of their expression levels and the incidence of DR was analyzed.Methods:The subjects were divided into three groups,healthy control Group A,simple diabetes Group B,diabetic retinopathy Group C.Three groups were determined the level of such indicators as,serum RhoA protein,intercellular adhesion molecule 1(ICAM-1),vascular cell adhesion molecule 1(VCAM-1);and we also determined the other related sugar metabolic indexes,fasting blood glucose(FBG),glycosylated hemoglobin(HbA1c),lipid metabolic indexes,total cholesterol(TC),triglyceride(TG),and the assessment indexes for liver,kidney function,like creatinine(CR),alanine aminotransferase(ALT)aspertate aminotransferase(AST).The content of RhoA protein,ICAM-1 and VCAM-1 in serum was determined by enzyme-linked immunosorbent assay(ELISA).Then calculated and analyzed the expression level and correlation of the above indexes.SPSS22.0 was used for statistical analysis and calculation.Result:1.Serum RhoA level: RhoA level in the DR group was significantly higher than that in the simple diabetes group(37.1±5.8pg/ml vs.31.1±6.3pg/ml,P<0.01).RhoA levels in the simple diabetes group and the DR group were significantly higher than those in the healthy control group(31.1±6.3pg/ml and 37.1±5.8pg/ml vs.23.2±6.08pg/ml,P<0.01).2.ICAM-1 level: the ICAM-1 level of the DR group was significantly higher than that of the simple diabetes group(373.8±82.6ng/ml vs.304.3±70.3ng/ml,P<0.05).ICAM-1 levels in the simple diabetes group and the DR group were significantly higher than those in the healthy control group(373.8± 82.6ng/ml and 304.3± 70.3ng/ml vs.212.5± 72.3ng/ml,P<0.01).3.VCAM-1 level: VCAM-1 level in the DR group was not significantly higher than that in the simple diabetes group(3.9±0.8ng/ml vs.3.8±0.7ng /ml,P>0.05).The VCAM-1 level in the simple diabetes group and the DR group was significantly higher than that in the healthy control group(3.8±0.7ng/ml and 3.9± 0.8ng/ml vs.3.21±0.88ng/ml,P<0.05).4.FBG and HbA1 c levels: FBG and HbA1 c levels in the simple diabetes group and the DR group were significantly higher than those in the healthy control group(10.15±4.09 and 11.59±4.71 vs.5.25±0.60,P<0.01;9.32±2.02 and 9.54±2.11 vs.5.46±0.34,P<0.01);There was no significant difference in FBG and HbA1 c between the DR group and the simple diabetes group(P>0.05).5.Serum RhoA level was significantly positively correlated with ICAM-1,VCAM-1,FBG,and HbA1c(the two indexes with the highest correlation coefficient were ICAM-1 and FBG,P<0.05).There was no correlation between serum RhoA level and age,gender,TC,TG,Cr,ALT and AST(P>0.05).6.The results of multiple linear regression analysis showed that serum RhoA expression was positively correlated with serum icam-1 expression(β=0.433,P<0.01)and FBG(β=0.266,P<0.05).Conclusion:1.There was a significant positive correlation between serum RhoA level and ICAM-1,VCAM-1,FBG,HbA1c(the two indexes with the highest correlation coefficient were ICAM-1 and FBG,P<0.05).2.Serum RhoA protein level was linearly correlated with ICAM-1 and FBG,suggesting that hyperglycemia may play an important role in the formation of DR by increasing the expression of RhoA protein to up-regulate the expression of ICAM-1. |
