Objectives: Breast cancer is the malignant tumors of mammary epithelium,and its incidence is the second highest in the world.Despite various treatment methods,recurrence and metastasis are major obstacles for BC treatments.The formation of a vascular network is extremely important for tumor growth and metastasis.It is already known that angiogenesis has a direct impact on BC development and that its level influences the prognostis of patients with this malignancy.Vascular endothelial growth factor(VEGF)is the most characteristic of angiogenic factor,it played an important role on the process of growth and metastasis in breast cancer.The level of VEGF in the tumor tissue can be used as an index for the diagnosis and prognosis of the tumor.This study detects the level of the density of ES,VEGF from organization and the concentration of PF-4 in serum,in order to find out the change rule of ES,VEGF and PF-4 in breast invasive ductal carcinoma patients.Then explore the relationship between them and other clinicopathological characteristics.Methods:1.The subject investigaated: 63 case of surgical treatment which were diagnosed as breast infiltrating ductal carcinoma were collected and prescreened from the CANGZHOU People's Hospital during July 2016 to July 2017.Select 30 case of breast benign lesions by the same method as control.Select 63 cases of patients,who had no received chemotherapy,radiotherapy,endocrine therapy and molecular targeted therapy,with no previous history of cancer patients.Take tumor tissue out from the tissue samples within 30 minutes in vitro,fixed by the concentration of 10% neutral formalin,and embedded in paraffin to be detected.The Preoperative fasting vein blood 2ml of all subjects were obtained to centrifuge by 1000r/min for 3 minutes,stored the upper serum in-80?.2.According to the results of immunohistochemistry,the correlation of expression of VEGF and ES,clinicopathological features and prognosis were analyzed respectively and reciprocally.3.ELISA method was applied to detect the serum levels of PF-4 in 63 breast invasive ductal carcinoma patients and 34 breast benign lesions.4.The statistical analysis: The analysis of data use the homogeneity of variance test by the SPSS 21.0 statistical software was according with normal distribution,Statistical methods using ?2 test,t test and correlation analysis.Result:1.The relationship between expression of VEGF and clinicopathologic features of IDCThe expression of VEGF was mainly found in the cytoplasm of tumor cells.49 cases of invasive ductal carcinoma and 8 cases of breast benign lesions were positive,the positive rate were 77.78%(49/63)and 23.53%(8/34)respectively,it's significantly higher than control group(P<0.05).There are positive correlation were found between the expression of VEGF and clinical pathological features,such as axillary lymph node metastasis,histological grade,Vascular invasion and Luminal typing(P<0.05),but no significant relationship were found with ages,tumor size and TNM stage(P>0.05).2.The relationship between expression of ES and clinicopathologic features of IDCThe expression of ES was mainly found in the cytoplasm of tumor cells.40 cases of invasive ductal carcinoma and 6 cases of breast benign lesions were positive,the positive rate were 63.49%(40/63)and 17.65%(6/34)respectively,it's significantly higher than control group(P<0.05).There are positive correlation were found between the expression of ES and clinical pathological features,such as axillary lymph node metastasis,TNM stage histological grade and Vascular invasion(P<0.05),but no significant relationship were found with ages,tumor size and luminal typing(P>0.05).3.The relationship between serum PF-4 and clinicopathologic features of IDCThe concentration of serum PF-4 in breast invasive ductal carcinoma patients were(0.3650.134)ng/ml,obviously higher than the control group which was(0.2110.089)ng/ml(P<0.05).There are positive correlation were found between the concentration of serum PF-4 and clinical pathological features,such as axillary lymph node metastasis,histological grade and Vascular invasion(P<0.05),but no significant relationship were found with ages,tumor size,TNM stage and luminal typing(P>0.05).4.The relationship between expression of VEGF,ES and PF-4 in invasive ductal carcinoma of breastThere was a significant correlation between VEGF and ES in breast invasive ductal carcinoma patients,it was a positive correlation,r=0.790,P<0.001.There was a significant correlation between serum PF-4 and VEGF expression in breast invasive ductal carcinoma patients,it wss a positive correlation,r=0.344,P=0.006.The levels of serum PF4 and ES expression had no correlation,r=0.092,P=0.475.Conclusion:1.In the IDC tissue,the expressions of VEGF and ES were significantly higher than those of the breast benign lesions.The concentration of serum PF-4 in IDC tissue were obviously higher than that in control group.2.The positively correlated with expression of VEGF and ES in breast invasive ductal carcinoma patients.3.The positively correlated with expression of serum PF-4 and VEGF in breast invasive ductal carcinoma patients.The expression of serum PF-4 had no correlation with ES.
