Objective This experiment to pyramidal tract preparation of anhydrous ethanol injection spastic paralysis rats model as the research object.To investigate the effects of the centripetal and centrifugal motion on the expression of soleus and gastrocnemius myosin heavy chain(Myosin heavy chain,MHC)subtype protein muscle of spastic paralysis rats.It provides theory basis for the rehabilitation of spastic paralysis which exercise is better in the future.Methods 30 newborn Sprague-Dawley rats at the 5th day of term were randomly divided into control group(on the left side of the pyramidal tract was injected of saline solution),model group(on the left side of the pyramidal tract was injected of anhydrous ethanol),the control group(on the left side of the pyramidal tract was injected of anhydrous alcohol,do not make any training),the centripetal movement group(on the left side of the pyramidal tract was injected of anhydrous ethanol,animals running on slope),the eccentric exercise group(on the left side of the pyramidal tract was injected of anhydrous ethanol,animals running on downhill),with 6 rats in each group.At the age of 20 days,for the control group and model group,we tested the two groups as follows: the behavioral manifestations of rats in each group were observed,the weight and muscle tension of rats in each group were detected.The pathological morphological changes of the pyramidal tract of brain tissue were observed by HE staining light microscope,and the expression of MHC-I and MHC-IIb proteins in the right soleus muscle and gastrointestinal muscle of rats in each group were detected by Western blotting.After 4 weeks of exercise training,the control group,eccentric exercise group and concentric exercise group were tested as follows: the weight and muscle tension of rats in each group were detected,the expression of MHC-I in soleus muscle,and MHC-IIb in gastriculus muscle.Results Compared with the sham operation group,the rats in the model group showed poor sucking activity two days after the operation to reduce the symptoms of convulsion,rolling and tremor and decreased response to external stimuli;At the age of 20 days,compared with the sham operation group,the rats in the model group walked slowly and limped in the circle,and showed increased muscle tone when the right limb was passive.At the age of 5 days,no abnormalities were observed in the sham operation group.At the age of 7 days,the weight of the model group was significantly lower than that of the sham group(P < 0.05).At the age of 20 days,the weight of the model group was significantly lower than that of the sham operation group(P<0.05).After exercise,the weight of each group was not statistically significant(P>0.05).The neuronal morphology of the sham operation group was normal under the light microscope,the nucleus was large and regular,and the nucleolus of the nuclear membrane was clearly visible compared with the sham operation group.After exercise,the improvement of muscle tone in the Eccentric exercise group was better than that in the Concentric exercise group(P<0.001).In spasm,MHC-I protein in skeletal muscle of rats is highly expressed and MHC-IIb protein is low expressed(P<0.001).Different exercise modes can increase the expression of MHC-IIb protein in gastrocnemius muscle to different degrees,and Eccentric exercise group > Concentric exercise group > control group(P<0.001).Different exercise modes can reduce the MHC-I protein expression of soleus muscle to different degrees.Eccentric exercise group > Concentric exercise group > control group(P<0.001).Conclusions The expression of MHC-I protein in skeletal muscle of spastic paralysis rats was dominant.Centrifugal motion can reduce muscle tension more than centripetal motion.Centrifugal exercise can lower the expression of MHC-I protein in soleus spasticity muscle and increase the expression of MHC-IIb protein in gastrocnemius muscle than centripetal exercise.
