Primary liver cancer(PLC),refers to malignant tumors occuring in hepatocytes or intraphepatic bile duct epithelial cells.It is one of the most common malignant tumors in China,which has the characteristics of high malignancy,rapid progression and being difficult to cure on average,about 250,000 people died of liver cancer peryear in the world,and about 45% of them are in China.The distribution of liver cancer incidence and death of hepatocellular carcinoma in China also has obvious regional characteristics.The high-incidence areas are mainly in the southeast coast,and Fujian province is a high-incidence area.The study is comprised of two parts:(1)retrospective analysis towards clinical data of hospitalized liver disease patients in southern Fujian province,which is separated into liver disease group and liver cancer group to study the epidemiological characteristics and related risk factors of primary liver cancer in southern Fujian province.Statistical analysis of clinical indicators was performed using R 3.4.2 software.Measurement data were expressed as mean and range,and t-test was used for comparison between groups.The count data is expressed as a percentage of the number of use cases,and the inter-group rate is compared using a chi-square test;(2)After the blood sample DNA is extracted using the kit,whole genome DNA methylation sequencing is performed to construct a single-base resolution DNA methylation expression profile in HBV liver cancer group and non-HBV liver cancer group,which can provide theoretical basis for the investigation about the pathogenesis of HBV-related liver cancer.The main results are as follows:(1)the results showed that the patients in the liver disease group were mainly 20 to 50 years old,and the patients in the liver cancer group were mainly 40 to 70 years old.There were more male patients than female patients in both cases,and more rural patients than urban patients.The factors such as Gender(male),age(?40 years),history of alcoholism,history of diabetes,cirrhosis,family history of liver cancer,and hepatitis B virus were significantly different between the two groups,which may be risk factors for PLC in southern Fujian province.(2)20464 DMRs were detected between HBV liver cancer group and non-HBV liver cancer group,corresponding to 8213 DMGs.Some significant enriched GOs related to tumor development were found by Gene Ontology functional enrichment(eg,cell development,cytoplasm,nucleoplasm,protein binding,etc.).Some significant enriched pathways related to tumor development were found by Kyoto Encyclopedia of Genes and Genomes pathway enrichment of DMGs(eg,ribosomes,adhesions).(3)in this study,many cancer-related gene loci that have been reported were rediscovered in the promotor CG,CHG and CHH sequence environment,and the proto-oncogene FYN,which may be associated with HBV hepatocellular carcinoma,has been excavated.Conclusion: this research is the first to explore the epidemiological characteristics of liver cancer in southern Fujian province,the results showed that gender(male),age(?40 years),history of alcoholism,history of diabetes,cirrhosis,family history of liver cancer,and hepatitis B virus may be risk factors for PLC in southern Fujian province.Genome-wide DNA methylation sequencing reveals the methylation differences between HBV hepatocellular carcinoma and non-HBV hepatocellular carcinoma,20464 DMRs were detected between HBV liver cancer group and non-HBV liver cancer group,corresponding to 8213 DMGs.Some significant enriched GOs related to tumor development were found by Gene Ontology functional enrichment.Some significant enriched pathways related to tumor development were found by Kyoto Encyclopedia of Genes and Genomes pathway enrichment of DMGs.The proto-oncogene FYN,which may be associated with HBV hepatocellular carcinoma,has been excavated.
